涎腺肿瘤中端粒长度及端粒酶活性测定的意义

摘    要:目的:探讨端粒长度及端粒活性变化在涎腺肿瘤发生中的作用和端粒酶活性水平与涎腺肿瘤生物学行为间的关系。方法:利用TRAP法分别测定35例良性涎腺肿瘤及24例恶性涎腺肿瘤的端粒酶活性,分析其表达情况及与涎腺肿瘤生物学行为间的关系。利用Southern杂交方法测定良,恶性涎腺肿瘤及正常涎腺的端粒长度,并进行统计分析。结果:良性肿瘤端粒酶表达率为17.14%,而恶性肿瘤表达率为90%,5例有触压痛的良性涎腺肿瘤及3例有面神经受累症状的恶性涎腺肿瘤患者均为端粒酶强阳性,35例良性肿瘤中,有32例端粒酶长度是缩短的,有2例长于正常涎腺端粒长度;24例恶性肿瘤中,有15例短于正常涎腺端粒长度,有5例长于正常涎腺端粒长度,有4例处于正常涎腺端粒长度范围内,15例短于正常涎腺端粒长度的恶性涎腺肿瘤均有酶表达,而32例良性肿瘤端粒酶活性表达率为10.34%,结论:端粒缩短并激活端粒酶是涎腺肿瘤形成的重要因素,端粒酶可能会成为涎腺恶性肿瘤的一个新的肿瘤标志物,恶性涎腺肿瘤不良的生物学行为与端粒酶活性水平间有高度相关性。

Abstract:Objective To probe the function from the change of telomere length and telomerase activity during the oncogenesis of salivary neoplasm and the correlation between telomerase activity level and the biological behavior of the salivary neoplasm.Methods The datum was derived from 35 cases of benigh and 24 cases of malignant salivary neoplasm.The activity in the benigh and the malignant neoplasm was detected by TRAP method respectively.The telomerase expression and the correlation between the telomerase expression level and the biological behavior of the neoplasm were analysed.The telomere lengths in benigh,malignant salivary neoplasm and normal salivary gland were measured by southern blot and were analysed statistically.Results The expression rate of benign salivary neoplasm was 17.14%;but 90% in malignant salivary neoplasm.5 cases of the malignant neoplasm with tenderness and 3 cases of the malignant neoplasm with facial nerve function damaged were all strong positive activity.In 35 cases of benigh salivary neoplasm,telomere lengths of 32 cases were reduced and the lengths of 2 cases were longer than the mean length of the normal salivary gland.In 24 cases of malignant salivary neoplasm,telomere lengths of 15 cases were shorter than the mean length of normal salivary gland,5 cases longer than the normal mean length and 4 cases within the normal mean length.The 15 cases malignant tumor with shorter telomere length were all strong positive activity.In 32 cases of the benigh tumor with shorter telomere length,the rate of telomerase activity was 10.34%.Conclusion Telomere reduction and telomerase activation are critical factor during oncogenesis.Telomerase could become a new marker of malignant tumor.There was an obvious correlation between telomerase activity level and the biological behavior of malignant salivary neoplasm.
