摘    要:目的:研究通心络胶囊对脑缺血大鼠脑细胞超微结构及脑线粒体呼吸功能的影响。方法:采用线栓法制作大鼠大脑中动脉缺血模型,透射电镜观察缺血后脑组织神经元超微结构的改变.检测线粒体内的NAD及FAD氧化呼吸链R3、R4、RCR、P/O等评价呼吸功能的指标,观察通心络(1.0、2.0mg/kg)对上述结构和指标的影响。结果:电镜照片显示通心络能改善脑组织神经元在缺血后的结构破坏情况;通心络治疗组大鼠线粒体呼吸功能明显改善。结论:通心络对缺血脑细胞有明显保护作用.机制与其能改善线粒体呼吸功能有关。

Abstract:Objective: To investigate the effects of Tongxinluo capsule on ischemic brain cell ultrastructure and mitochondrial respiratory function in rats. Methods: The models of middle cerebral artery occlusion (MCAO) rats were produced by Zea Longa method. Transmission electron microscopy (TEM) was used to observe the changes of brain cell ultrastructure and R3, R4,RCR and P/O of mitochondrial NAD chain or FAD chain were tested to estimate mitochondrial respiratory function. Results: The result of TEM examination showed that the ultrastructure destruction was reduced by Tongxinluo (1. 0, 2.0 mg/kg). Mitochondrial respiratory function was significantly improved in Tongxinluo (1.0. 2.0 mg/kg) groups. Conclusion: Tongxinluo showed a protective effect on ischemic cerebral cells. The mechanism may be related to improvement of mitochondrial respiratory function.
