原发性高血压与多巴胺受体D1基因多态性的相关性研究

摘    要:目的 探讨多巴胺受体D1基因 4 8A/G多态性与原发性高血压 (PH)的相关性。方法 运用聚合酶链反应—限制性片段长度多态性分析 (PCR RFLP)方法 ,了解 - 48A/G基因型在PH组和正常血压对照 (NT)组的分布情况。结果 等位基因A、G在PH组和NT组的分布频率分别为 0 .78、0 .2 2和 0 .86、0 .14,基因频率分布符合Hardy Weinderg平衡 ,样本具有群体代表性 ,两组人群的基因型和等位基因频率存在明显统计学差异 (P <0 .0 1,P <0 .0 1)。PH组中G等位基因、舒张压明显高于A等位基因 (P <0 .0 5 )。结论 本组人群中 ,多巴胺受体D1基因 -48A/G多态性与PH显著性相关。

Abstract:Objective The present study examined the association between dopamine D 1 receptor gene-48A/G polymorphism and primary hypertension in Hubei province.Methods The distribution of -48A/G polymorphism between primary hypertensive patients and normotensive subjects was identified with polymerase chain reaction-based restriction fragment length polymorphism protocol.Results The allelic frequencies were 0.78 for the A allele and 0.22 for the G allele in PH patients,0.86 for the A allele and 0.14 for the G allele in normotensive subjects.The allelic frequencies were coincident with Hardy-Weinberg equilibrium.Samples were of popular representative. Significant differences in the genotype distribution (P<0.01) and allelic frequencies (P<0.01) between hypertensive and normotensive subjects were seen,and the diastolic blood pressure for Gallele was higher significantly than that for allele in hypertensive subjects (P<0.05).Conclusion These results suggest that the -48A/G polymorphism of human dopamine D 1 receptor associates significantly with primary hypertension in this population.;
