化疗药物对培养状态下脊索瘤细胞生长的抑制作用

目的：采用细胞培养技术及ＭＴＴ法，观察培养状态２下脊索瘤细胞的生长特点及化疗药物对脊索瘤生长的抑制作用。方法：手术切除的新鲜脊索瘤标本，置入含１００ｍＬ／Ｌ小牛血清的ＲＰＭＩ１６４０培养基中，观察脊索瘤细胞生长特点；取对数生长期的细胞接种９６孔板，每孔细胞数１０＾４个，将质量浓度３，３０，７５ｍｇ／Ｌ顺铂；１５，１５０，３７５ｍｇ／Ｌ卡铂；１，１０，２５ｍｇ／Ｌ表阿霉素分别加入孔内，３７℃５０ｍＬ

Inhibitory effects of anti-cancer chemical drugs on cultured chordoma cells

AIM: To observe the growth feature of chordomace1ls under the culture conditions and the growth inhibitoryeffects by chemotherapeutic drugs on the cultured ce1ls.METHODS: The resected chordoma tissues, which were firstcut into 13 mm in size and washed with serum free RPMI164O medium, were cu1tured in complete RPMI 164O medi-um. The chordoma cells from the cultured tissues wereobserved under microscope several days later, counted andseeded in the 96 well plate with 1O' cells per well when theywere in their logarithmic growth phase. Three differentchemotherapeutic drugs at different mass concentrations wereadded into the 96 well plate with sisplatin at 3, 3O, 75 mg/Land carborp1tin at l5, 15O, 375 mg/I, and epirubicin hy-.drochloride at l, 1O, 25 mg/L respectively and cultured for48 hours. At the end of the culture, cell growth inhibitionrates by the drugs were calculated by using MTT method.RESULTS: The morpho1ogy of cu1tured chordoma ce1ls fromresected tissues were of different sizes and different shapes.Adhesion growth and mitogenesis were easy to see. The inhi-bition rates of chemotherapeutic drugs, sisplatin and carbor-pltin, were almost the same (9%~ 22% ) and this effect increased with the increasing mass concentrations of the drugs.But this effect was not observed in epirubicin hydrochloride.CONCLUSION: Chemotherapeutic drugs sisplatin and Car-borpltin but not epirubicin hydrochloride have inhibitory ef-fects on the chordoma cell growth.
