复肝春6号抗H22肝癌作用机制的实验研究

摘    要:目的探讨复肝春6号(FGC-6)抗肝癌作用机制。方法建立荷H笠肝癌小鼠模型,以DNA琼脂糖凝胶电泳观察FGC-6对肿瘤细胞诱导凋亡的作用,以Western blot检测肿瘤组织中bcl-2、bax基因的蛋白表达水平;制备FGC-6含药血清,观察其对体外培养H22细胞的增殖抑制作用。结果FGC-6可诱导肿瘤组织DNA片段化,琼脂糖凝胶电泳结果可见梯状条带出现;Western blot结果显示FGC-6使肿瘤组织中bcl-2基因表达量明显降低,而bax基因的表达水平却明显升高;FGC-6含药血清对体外培养H22细胞增殖有明显抑制作用。结论FGC-6的抗肝癌作用可能与调节bcl-2和bax基因的表达量,诱导肝癌细胞凋亡有关。

Abstract:Objective To explore the mechanism on anti-hepatic carcinoma of Fuganchun 6(FGC-6).Methods The model mice bearing H_(22) hepatic carcinoma were established to observe the apoptosis of carcinoma cells induced by FGC-6 by DNA agarose gel electrophoresis and to detect the protein expression of bcl-2 and bax gene in carcinoma tissue by Western blotting.The inhibitive effects of serum containing FGC-6 in H_(22) cells on H_(22) cells were studied in vitro.Results FGC6 induced the DNA of tumor cell ruptured into segments having ladder lanes in the results of agarose gel electrophoresis and reduced the level of bcl-2 protein of the tumor cells,while increased the level of bax protein by Western blotting.The serum containing FGC-6 restrained the proliferation activity of H_(22) cells in vitro.Conclusions The mechanism of FGC-6 on antitumor might be related to the regualted level of bcl-2 and bax protein and the induced hepatic cells apoptosis.
