摘    要:研究大鼠大脑中动脉(MCA)短暂闭塞后血管源性脑水肿(VBE)与调亡细胞数的关系。VBE程度用光密度法测量甲酸胺提取外渗至血脑屏障(BBB)受损最严重的脑区Evans蓝(EB)含量定量,石蜡切片的调亡细胞数用原位末端标记法(ISEL)检测。结果表明BBB受损程度与调亡细胞数随缺血时间延长而增加二者密切相关。VBE促进了细胞凋亡机制。

Abstract:To evaluate the relation between the degree of vasogenic brain edema (VBE) and the unmber of apoptotic cells following temporary focal cerebral ischemia. The degree of VBE produced was assessed by quantitation of the extravasation of Evans blue dye (EB) into the area of maximum bloodbrain barrier (BBB) breakdown by means of optical densitometry following formamide extraction. Apoptiotic cells were measured in paraffin sections with in situ end-labeling (ISEL) method. Both the extravasation of EB and the number of apoptotic cells increased with duration of focal cerebral ischemia. The extravasation of EB in the isohemic hemisphere correlated well with the number of apoptotic cells in the coronal section. It is indicated that vasogenic brain edema should be invoved in the mechanism of apoptosis.
