摘    要:目的:采用响应面法优化美洲大蠊中氨基酸的提取工艺参数。方法:在单因素实验的基础上,利用Box-Behnken中心组合设计,以氨基酸含量、干膏率为评价指标,对提取时间、料液比、醇提浓度、提取次数等4个主要影响因素进行考察,并进行工艺优化。结果:通过Box-Behnken效应面法及总评归一值(Optical Density,OD)系统评价,确定最优提取工艺如下:取饮片适量,加入12倍量70%乙醇溶液,提取2次,每次120min。利用该工艺条件得到的氨基酸含量及干膏率与理论预测值基本一致。结论:实验确立的响应面模型与实际情况拟合良好,能较好的预测美洲大蠊中氨基酸含量及干膏率等指标,该方法具有一定的实用价值。

Abstract:This study aimed to optimize the extraction process of free amino acid in Periplaneta americana L.by Box-Behnken design-response surface methodology.On the basis of single factor screening,a three-factor and three-level Box-Behnken experimental design was employed with extraction time(X1),solvent ratio(X2)and solvent concentration(X3)as independent variables.And extraction frequence was also involved in a major impact.Dependent variables were transformed into desirability mathematically by comprehensive evaluation method and Hassan''s method.Data of overall desirability was fitted to a second order polynomial equation,through which three dimensional response surface graphs were produced. Optimum experimental conditions were selected from the stationary point of the response surfaces.Through the Box-Behnken design-response surface methodology and prognostic scoring system optimized scheme,the preferred processing conditions were as follows:added 12times of 70% ethyl alcohol,extracted twice with each for 120min.The content of amino acid and the dry extra yield actually extracted were in coordinence with the disrable values.In conclusion,the extraction technology of free amino acid optimized by Box-Behnken Design was practical and feasible with good fitting degree for predicting indexes(e.g.free amino acid and dry extra yield)in Periplaneta americana L..
