天泰1号对自发老年性痴呆模型学习记忆及神经元NOS的影响

摘    要:目的观察天泰1号对自发老年性痴呆模型学习记忆功能及神经信使分子NO代谢的影响.方法从21月龄昆明种小鼠筛选出自发老年性痴呆(记忆障碍)鼠,随机分为4组,即空白对照组,西药对照组,补肝养髓小剂量组,补肝养髓大剂量组,另设老年学习记忆正常组(简称老年正常组,下同).西药对照组给以喜得镇(Hydergine)0.6 mg/kg,补肝养髓小、大剂量组分别予补肝养髓方6.80 g/kg及20.41 g/kg,连续60 d,正常对照组和痴呆对照组均灌以等量双蒸馏水(DW);用跳台实验(step-down test)检测学习记忆成绩;脑组织冰冻切片,NBT组织化学法示大脑NOS1活性,全自动显微图像分析系统定量.结果天泰1号能显著改善自发老年性痴呆模型小鼠的学习记忆障碍,并明显增强中枢大脑皮质和海马NOS1活性及其纤维密度,其作用呈现出一定的量效关系.此外,研究还发现颞叶皮质、海马NOS1活性与学习记忆成绩呈显著性正相关.结论中枢NOS1活性与学习记忆成绩显著相关;补肝养髓方可明显改善自发老年性痴呆模型的学习记忆障碍,这一作用可能与增强其大脑皮质和海马NOS1活性,调节中枢神经信使分子NO代谢有关.

Abstract:Objective To observe the effects of Tiantai No.1 formula on the cognitive ability and metabolism of nerve messenger molecule NO in spontaneous aged dementia mice.Methods Spontaneous aged dementia mice were screened out from 21 months old Kunming mice and then divided randomly into 4 groups: control, western medicine, lower dosage Tiantai No.1 and higher dosage Tiantai No.1 group, in addition, normal aged mice group was also set. Western medicine group was treated with Hydergine 0.6 mg/kg,while Traditional Chinese Medicine was given with the dosage of 6.80 g/kg and 20.41 g/kg respectively in lower dosage and higher dosage Tiantai No.1 group, control and normal aged groups were treated with equal volume DW. All groups were treated as aforementioned for 60 days, using step-down test to estimate the ability of learning and memory. Brain tissue was made to frozen section. Using NBT histochemical technique to measure the activity of NOS 1 and using automatic micro-image analysis system to determine quantity.Results Tiantai No.1 could improve learning and memory in spontaneous aged dementia mice remarkably, moreover, could enhance the activity and fiber density of NOS 1 in cerebra cortex and hippocampus. All these effects had a dose-response relationship. Furthermore, the results show the activity of NOS 1 in temporal lobe cortex and hippocampus had close positive correlation with the ability of learning and memory.Conclusion Activity of NOS 1 in CNS had close correlation with the ability of learning and memory. Tiantai No.1 could improve learning and memory in spontaneous aged dementia mice significantly, this effect might relate to its action of enhancing the activity of NOS 1 in cerebra cortex and hippocampus, regulating the metabolism of nerve messenger Molecule NO.
