高血糖对实验性糖尿病大鼠心脏血管病变的作用

摘    要: 目的研究高血糖在糖尿病大鼠心脏血管病变发生发展中的作用.方法链脲佐菌素在腹腔内注射制备糖尿病大鼠动物模型,免疫组化及电镜分别观察正常对照组(A组)、胰岛素治疗组(B组)、糖尿病未治疗组(C组)心血管病变.结果(1)16周时与A组及B组比较,C组体重显著减小、血糖及糖化血红蛋白(GHbAlc)显著增高、高密度脂蛋白胆固醇(HDL·c)显著降低.(2)免疫组化结果:16周时与A组及B组比较,C组actin、desmin阳性细胞计数显著减少,F8、VEGF阳性细胞计数显著增加.(3)电镜结果:16周时C组大鼠可见典型的心脏动脉粥样硬化样改变.其他两组则未见明显上述病变.结论高血糖是实验大鼠心脏动脉粥样硬化样病变发生发展的强烈危险因素.

Abstract:Objective To investigate the effect of high blood glucose on the occurrence and development of cardiovascular diseases in diabetic rats.Methods The diabetic rat model was developed by intraperitoneal injection of streptozocin.The cardiovascular diseases in normal control group (A),insulin-treated diabetic group (B),and diabetic group without treatment (C) were observed by immunohistochemical and electromicroscopic methods.Results (1)The weight of group C was significantly lower than those of groups A and B ( P < 0.05) ,and the blood glucose and glycosylated hemoglobin(GHbAlc) levels were also higher( P < 0.05) ,but the high-density lipoprotein cholesterol level (HDL-c)was significantly lower than in groups A and B ( P < 0.05) .There were no significant differencess ( P > 0.05) in plasma triglyc-erides (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-c) and mean arterial pressure between the groups. (2) Immunohistochemical analysis showed that the of actin and desmin positive cells in group C were significantly less than those in groups A and B, but F8 and VEGF positive cells were significantly increased( P <0.05).(3)In group C the typical atherosclerotic changes observed under electron microscope, while no typical changes were seen in the other two groups. Conclusion Hyperglycemiais a high risk factor in the occurrence and development of cardiovascular diseases in experimental diabetic rats.
