白细胞介素-6对破骨细胞骨吸收效应及MMP-3表达影响的实验研究

摘    要:目的:观察不同浓度IL-6对破骨细胞骨吸收的剂量效应及对破骨细胞基质金属蛋白酶-3表达的影响,以期进一步阐明IL-6介导基质金属蛋白酶在破骨细胞性骨吸收中的病理机制。方法:采用体外破骨细胞溶骨模型,通过原子吸收分光光度仪及免疫组化染色技术检测不同浓度IL-6对破骨细胞溶骨活性及其质金属蛋白酶-3表达的影响。结果:当IL-6>10U/ml时,培养上清中Ca^2 浓度显著增加,牙本质片上骨吸收陷窝数目明显增多;当IL-6浓度为100U/ml、500U/ml时破骨细胞基质金属蛋白酶-3表达的阳性信号显著增强。结论:在IL-6作用下,破骨细胞表达基质金属蛋白酶-3。IL-6对破骨细胞具有激活作用,低浓度主要诱导破骨细胞形成,较高浓度刺激破骨细胞的溶骨活性。

Abstract:Objective:To explore the effect of IL-6 on the bone resorption activity and the expression of MMP-3 (matrix metalloproteinase-3) in osteoclasts,for further to understand the mechanisms of osteoclastic bone resorption regulated by IL-6.Methods:Set up a model of bone resorption in vitro,to investigate the influence of varying concentrations of IL-6 on the number of bone resorption pits and the release of valcium-used as parameters of bone resorption.The effects of IL-6 on the expression of MMP-3 in osteoclasts was examined by immunohistochemical technique.Results:The addition of IL-6 (10-500U/ml) induced a dose-dependent increases in the release of calcium and the number of bone resorption pits.When the concentration of IL-6 was 100U/ml or 500U/ml,the expression of MMP-3 in osteoclasts increased significantly over those cells treated with 10U/ml.Conclusions:The bone resorption activity and the expression of MMP-3 in osteoclasts appear to be mediated by IL-6.Lower concentration of IL-6(10U/ml) mainly induced the formation of osteoclasts in rat marrow cells culture,and higher concentration (100U/ml or 500U/ml) stimulate the bone resorption activity through increased the expression of MMP-3 in osteoclasts.
