摘    要:目的探讨阿托伐他汀对血管新生的促进作用及其作用机制。方法建立野生型C3H/He小鼠下肢缺血模型,观察用药后肢体血流、毛细血管数、血管内皮生长因子(VEGF)蛋白表达。并行离体实验(血管新生共同培养),计数血管样结构物管腔形成数。结果阿托伐他汀使实验小鼠术后缺血肢血流明显改善,缺血肢与非缺血肢血流面积比明显增加;缺血肢毛细血管密度明显增加,VEGF蛋白表达增强。血管新生共同培养,血管样结构物管腔数明显高于对照组和加入血管新生抑制剂组,但与阳性对照组比较无明显差异。结论阿托伐他汀有促进血管新生的作用。

Abstract:Objective To study the promoting effect of atorvastatin on angiogenesis.Methods The model of hind- limb blood of C3H/He mice was established.Hind- limb blood flow was measured by Laser Doppler perfusion imager (LDPI) before treatment and after ischemic operation of the hind-limbs. The number of capillaries was measured by immunofluorescence technique. The expression of vascular endothelial growth factors (VEGFs) in the ischemic limbs was observed by Western blot.The number of vessel-like structures in each hole was calculated and compared with VEGF (positive groups) and suramin (neo-angiogenesis inhibitor) by microscope. Results After treated with atorvastatin, the blood flow of the ischemic limbs recovered obviously. The ratio of blood flow area between the ischemic limbs and non-ischemic limbs was greatly increased. Atorvastatin increased the density of capillaries in the ischemic limbs of C3H/He mice. Atorvastatin enhanced the expression of VEGF in the ischemic limbs of C3H/He mice.After co-culture of neo-angiogenesis,The number of vessel-like structures was significantly higher in atorvastatin group than that in both control and neo-angiogenesis inhibitor groups,which was not significantly different between atorvastatin group and VEGF positive control group. Conclusion Atorvastatin promotes the angiogenesis in vivo and in vitro in rats.
