野金柴抗炎、镇痛和免疫调节作用的研究

摘    要:目的研究野金柴的抗炎、镇痛、免疫调节作用。方法采用小鼠耳郭肿胀法、腹腔毛细血管通透性观察法、扭体法、碳粒廓清法和迟发型超敏反应法研究野金柴水提物和醇提物的抗炎、镇痛、免疫调节作用。将小鼠随机分8组,分别为空白、阳性对照、野金柴水提取物(低、中、高剂量)、野金柴醇提取物(低、中、高剂量)组。实验结束时测定相应指标。结果野金柴水提物高剂量和醇提物中剂量均能显著减轻二甲苯致炎小鼠的耳肿胀度(P〈0.05或P〈0.01);水提物低、中、高3个剂量均能显著抑制对乙酸所致腹腔毛细血管通透性的增大(P〈0.05或P〈0.01);水提物及醇提物均能显著抑制乙酸所致扭体次数(P〈0.01);水提物高剂量和醇提物中剂量能显著提高小鼠吞噬细胞的吞噬功能(P〈0.05或P〈0.01);除醇提物低剂量外,水提物及醇提物各剂量均能显著抑制过敏反应引起的耳肿胀(P〈0.05或P〈0.01)。结论野金柴具有一定的抗炎、镇痛作用,对免疫功能有一定的双向调节作用。

Abstract:Objective To explore the anti-inflammatory,analgesic effects and immunologic function of the ethanol extract and aqueous extract of Lithocarpus polystachyus Rehd.leaves(LP) in mice.Methods Dimethylbenzene-induced ear edema method and acetic acid-induced capillary permeability method were used to observe the anti-inflammatory effects of LP;acetic acid-induced writhing test in mice was performed to observe the analgesic effects of LP;carbon clearance method and DNCB-induced delayed type hypersensitivity(DTH) method was used to evaluate the effects of ethanol extract and aqueous extract of LP on immunologic functions.Mice were randomly divided into eight groups,i.e.,normal control group,positive control group,three groups treated with aqueous extracts of LP at high,intermediate,and low doses respectively,and three groups treated with ethanol extract of LP at high,intermediate,and low doses respectively.Results Dimethylbenzene-induced ear edema in mice could be significantly inhibited by aqueous extract of LP at high dose and ethanol extract of LP at intermediate dose(P0.05 or P0.01).Increased peritoneal capillary permeability in mice induced by acetic acid was significantly inhibited by aqueous and ethanol extracts of LP respectively at high,intermediate,and low doses(P0.05 or P0.01).The number of writhing in mice was significantly inhibited by aqueous and ethanol extracts of LP respectively(P0.01).And phagocytosis of phagocytes of mice was be significantly improved by aqueous in mice caused by hypersensitive reaextract of LP at high dose and ethanol extract at intermediate dose(P0.05 or P0.01).And ear edemactions was significantly inhibited by aqueous and ethanol extracts of LP at different doses except ethanol extract of LP at low dose(P0.05 or P0.01).Conclusion Aqueous extract and ethanol extract of LP had anti-inflammatory,analgesic effects and two-way immunoregulative effects to some extent.
