感染乳鼠组织中克里米亚-刚果出血热病毒的RT-PCR检测与形态学鉴定

目的：检测并鉴定感染组织中的克里米亚-刚果出血热病毒。方法：分别经皮下与腹腔两种途径感染乳小鼠；应用RT-PCR技术对病毒核酸进行检测；利用透射电镜超薄切片技术对病毒进行形态学鉴定。结果：RT-PCR可在心脏、肝脏、脾脏、肾脏以及脑组织内检测到病毒核酸，透射电镜下可在肝细胞胞浆内观察到形态典型的病毒粒子。结论：应用RT-PCR技术可以对感染组织中的克里米亚-刚果出血热病毒进行检测，利用透射电镜可以对病毒的形态进行观察与鉴定。

RT-PCR detection and morphological identification of CCHF virus in infected suckling mice

Objective:To detect and identify Crim ean-Congo hemorrhagic fever(CCHF) virus in infected tissues.M ethod s:The suckling m ice were infected w ith CCHF virus by subcutaneous or intraperitoneal in jection,and CCHF virus in the infected tissues was exam ined by RT-PCR.Morphology of CCHF virus was identified by transm ission electron m icrosco-py.R esu lts:The nuc le ic ac id of CCHF virus cou ld be detected in the hearts,livers,sp leens,k idneys and brains of the in-fected suckling m ice by the RT-PCR.V irus w ith typ ical configuration cou ld be found in the cytop lasm of the hepatocytes under the transm ission electron m icroscope.C onc lu sion:The virus in infected tissues can be detected by RT-PCR and morphology of CCHF virus can be identified by transm ission electron m icroscopy.
