抗T细胞单克隆抗体对再生障碍性贫血患者TNF和IL—2水平的影响

为探讨抗Ｔ淋巴细胞克隆抗体对再生障碍性贫血患者免疫功能的调节作用，采用放射免疫检测２５例ＡＡ患者ＭｃＡｂ－Ｔ治疗前后血清肿瘤坏死因子和白细胞介素－２（ＩＬ－２）水平及其中１０例周围血单个核细胞体外诱生ＴＮＦ和ＩＬ－２水平的变化。

THE EFFECT OF ANTI HUMAN T LYMPHOCYTE MONOCLONAL ANTIBODY ON TNF AND IL 2 LEVEL IN APLASTIC ANEMIA PATIENTS

The aim of this study was to investigate the effect of immunoregulatory therapy with monoclonal anti human T lymphocyte antibody(McAb T) on patient with aplastic anemia (AA).The level of tumor necrosis factor (TNF) and interleukin 2(IL 2) both in serum of 25 patients and in supernatant of phytohemagglutinin(PHA ) cocultured peripheral blood mononuclear cells(PBMNC) culture from 10 patients with AA were analysed by using radioimmunoassay methods before and after McAb T therapy. The results showed that before therapy both TNF in serum and TNF or IL 2 in culture supernatant of PBMNC in effective group were significantly increased than normal control ( P <0.01),but they were all decreased and showed no difference compared with normal control ( P > 0.05 ) after therapy. In ineffective group,the levels of TNF and IL 2,both in serum and in culture supernatant,showed no significant difference before and after therapy,but the TNF and IL 2 levels in culture supernatant were both higher than that in normal control ( P < 0.05) before and after therapy.The results suggested that the hematopoietic regulation cytokines were abnormal in AA patients,McAb T could play a specific immunoregulatory role for hematopoietic regulation cytokines including TNF, IL 2.
