摘    要:目的:优化小鼠雪旺细胞(SCs)的离体培养方法。方法:取新生小鼠的背根神经节,采用酶消化分散法,加入不同的培养液,第1组(NB组)用NB(neuronbasal medium),第2组(FBS组)用含10%FBS的DMEM,继用O4、S-100免疫荧光法染色鉴定SCs阳性细胞,并镜下计数,计算SCs纯度。结果:NB组SCs纯度于第1周为90%,第2周为86.6%;FBS组相应SCs纯度都低于30%。结论:无血清的NB培养液更有利于SCs的增殖和生长。

Abstract:Objective:The method of culturing Schwann cells(SCs) from the dorsal root ganglia of mice has been optimized.Methods: SCs were dissociated from the dorsal root ganglia(DRGs) in 1-day-old mice,fed with one of two kind media: Neurobasal media without serum,or DMEM plus 10% FBS.The purity of the SCs was evaluated by percentage of O4 and S-100-immunostaining cells.Results: The purity of SCs in cultures fed with NB reached up to 86%-90%,while the yield of purity fed with 10% FBS was below 30%.Conclusion: Neurobasal media without serum,used for neuronal cultures,is also favorable for SCs growth and/or proliferation in vitro.
