艾灸对胃黏膜损伤大鼠胃黏膜表皮生长因子、转化生长因子-α及其受体的影响

摘    要:目的:观察艾灸对应激性胃黏膜损伤大鼠胃黏膜表皮生长因子(EGF)、转化生长因子-α(TGF-α)与表皮生长因子受体(EGFR)的影响,探讨艾灸修复胃黏膜损伤的作用机制。方法:SD大鼠随机分为正常组、模型组、艾灸组,每组12只。束缚冷应激法制作应激性胃黏膜损伤大鼠模型,在造模之前,艾灸组选取"足三里""中脘""脾俞""胃俞"行艾灸预处理8d。酶联免疫法检测胃黏膜组织中EGF、TGF-α的含量,免疫组化法检测胃黏膜EGFR蛋白表达。结果:与正常组比较,模型组大鼠胃黏膜TGF-α的含量明显增加(P<0.05);与模型组比较,艾灸组大鼠胃黏膜EGF、TGF-α的含量及EGFR蛋白表达明显上调(P<0.05,P<0.01)。结论:预先艾灸"足三里""中脘""脾俞""胃俞"可上调应激性胃黏膜损伤大鼠胃黏膜EGF、TGF-α的含量及EGFR表达,可能与其对胃黏膜的保护作用有关。

Abstract:Objective To observe the effects of moxibustion on contents of epidermal growth factor(EGF) and transforming growth factor-alpha(TGF-α) and epidermal growth factor receptor(EGFR) expression in the gastric mucosa tissue in rats with gastric mucosal lesion.Methods Thirty-six SD rats were randomly and equally divided into control,model and moxibustion groups.Gastric mucosal lesion model was duplicated by restraint and cool water immersion stress.Pre-moxibustion was applied to "Zusanli"(ST36) and "Zhongwan"(CV12),"Pishu"(BL20) and "Weishu"(BL21) alternately,once everyday for 8 days before modeling.The contents of EGF and TGF-α in gastric mucosa were detected by enzyme-linked immunosorbent assay(ELISA) and the expression of EGFR determined by immunohistochemistry.Results Compared with the control group,only TGF-α content in the gastric mucosa in the model group was increased significantly(P<0.05).Compared with the model group,the EGF and TGF-α contents and EGFR immunoactivity in the gastric mucosa were increased significantly in the moxibustion group(P<0.05,P<0.01).Conclusion Pre-moxibustion at ST36,CV12,BL20 and BL21 can up-regulate gastric mucosal EGF and TGF-α contents and EGFR protein expression in gastric mucosa lesion rats,which may contribute to its effect in relieving stress-induced gastric mucosal injury.
