大鼠失血性休克与复苏后回肠组织凋亡的机制

摘    要:目的探讨大鼠失血性休克与复苏后回肠组织凋亡的可能机制。方法采用Chaudry方法(略作改动)制作大鼠失血性休克模型,60min后用回输血液和生理盐水进行复苏。检测不同的时相回肠组织Bax、Bcl-2、Bcl-2/Bax及Caspase-3蛋白的表达水平和电镜下回肠组织超微结构的改变。结果大鼠失血性休克与复苏后回肠组织Bax、Caspase-3蛋白表达上调,Bcl-2蛋白表达下调,Bcl-2/Bax比值下降,回肠组织凋亡明显。结论大鼠失血性休克与复苏后回肠组织凋亡的可能机制是上调回肠组织Bax蛋白和Caspase3蛋白的表达,下调Bcl-2蛋白的表达,使Bcl-2/Bax比值下降。

Abstract:Objective To investigate the probable mechanism of apoptosis of ileal tissue in rats with hemorrhagic shock and reperfusion. Methods The model of hemorrhagic shock and reperfusion in rats was produced by modified Chaudry method. After 60 minutes, rats were reperfusecl by transfusion of shed blood and normal saline . At different time points after reperfusion, the expression of Bax, Bcl-2 , Bcl-2/Bax , Caspase-3 in ileum tissue was measured using immunohistochemical method and the changes of ultramicroscopic structures of ileal tissue under electron microscope. Results The expression levels of Bax and Caspase-3 increased but Bcl-2 and the ratio of Bcl-2/Bax decreased after hemorrhagic shock and reperfusion. The apoptosis of ileum was significant. Conclusion The probable mechanism of apoptosis of ileal tissue in rats with hemorrhagic shock and reperfusion was not only upregulation of Bax and Caspase-3 but also clownregulation of Bcl-2 and the rate of Bcl-2/Bax.
