补肾通络方对去势大鼠阴茎组织NOS基因表达的影响

摘    要:目的 探讨一氧化氮合酶 (NOS)基因表达与补肾通络方在阴茎勃起中的关系。方法 将 4 0只成年雄性大白鼠随机分为对照组、去势组、补肾通络方组及丙酸睾酮组。去势组、补肾通络方组与丙酸睾酮组分别切除双侧睾丸 ,其中补肾通络方组和丙酸睾酮组在切除睾丸后给予中药及睾酮替代 ,于术后 3周取阴茎组织 ,用PCR仪检测其NOSmRNA的表达。结果 去势组NOSmRNA的表达较其他 3组下降 ;而表现NOSmRNA增多的组 ,由多到少排序为对照组、丙酸睾酮组及补肾通络方组 ;对照组、丙酸睾酮组及补肾通络方组其NOSmRNA内控量较多 ,与去势组比较有显著性差异(P <0 0 5 ) ,而丙酸睾酮组及补肾通络方组之间无显著性差异 (P >0 0 5 )。结论 补肾通络方可能通过调节NOSmRNA表达而在阴茎勃起中发挥重要作用。

Abstract:Objective To find the relationship between the gene expression of NOS and the effects of Bushentongluo Formula (BF) on the erection of penis. Methods 40 Adult rats were randomly divided into the control, castrated, BF, and testosterone propionate (TP) groups, and the testes of the rats in the last three groups were removed. BF group was given BF and TP group was given TP. Three weeks after the orchiectomy, the penis tissues of the rats in BF group and TP group were taken for detecting the expression of NOS mRNA by using PCR. Results The expression of NOS mRNA in the rats in the castrated group was lower than that in the other three groups. The order of the groups in which the expression of NOS mRNA showed an increase was: the control group > TP group > BR group. The internally controlled amounts of NOS mRNA in the control group, TP group and BF group were larger than those in the castrated group ( P <0 05), but there was no marked difference between the internally controlled amounts of NOS mRNA in TP group and BF group ( P >0 05). Conclusion BF can exert an important role in the erection of penis through regulating the expression of NOS mRNA.
