摘    要:目的:检测端粒酶的反义寡核苷酸对喉癌细胞的影响。Hep-2方法:采用法及平板集落形成实验检测靶向于端MTT粒酶的反义寡核苷酸对喉癌细胞株增殖的抑制作用。用法检测反义寡核苷酸对喉癌细胞株细胞内端粒Hep-2 PCR-ELISAHep-2酶活性的影响。结果:反义寡核苷酸明显抑制了喉癌细胞的增殖,此作用有剂量、时间依赖性。反义寡核苷酸可抑制喉Hep-2癌细胞端粒酶活性,此作用有浓度依赖性。结论:靶向于端粒酶的反义寡核苷酸能抑制喉癌细胞的增殖。Hep-2

Abstract:Objective: To investigate the effect of the antisense oligodeoxynucleotide (AS-ODN) targeting against telomerase on laryngeal carcinoma cells Hep-2 strain in vitro. Methods: MTT test and plat clone forming test were used to measure the proliferation inhibition of the AS-ODN targeting against telomerase on Hep-2 cell strain. Telomerase activity of Hep-2 cell strain was analysed by PCR-ELISA technique. Results: The AS-ODN inhibited Hep-2 cell strain obviously with dose dependence and time dependence. The AS-ODN inhibited telomerase activity of Hep-2 cell strain with density dependence. Conclusion: The AS-ODN targeting against telomerase could inhibit the proliferation of Hep-2 cell strain.
