肿瘤坏死因子-α-转化酶和EGFR在非小细胞肺癌转移中的作用研究

摘    要:目的探讨非小细胞肺癌(NSCLC)病理组织中肿瘤坏死因子-α-转化酶(TACE)和EGFR在原发灶和转移灶中的表达及其与患者预后的关系。方法应用液相芯片技术、RT-PCR、WesternBlot等方法测定57例NSCLC和14例肺部良性病变病理切片中膜结合肿瘤坏死因子(M—TNF)、TACE、EGFR的水平,对比分析TACE、EGFR在NSCLC患者和正常人之间以及NSCLC原发灶和转移病灶的表达。结果57例NSCLC中M—TNF和TACE的异常表达率分别为66.67%和64.91%,EGFR的阳性表达率为77.19%;14例肺部良性病变均未见TACE的过度表达:TACE和EGFR在NSCLC转移灶中的表达明显高于原发灶,并与预后成正相关。结论检测TACE和EGFR在NSCLC中的表达变化是判断恶性肿瘤程度、浸润转移的有效参考指标,TACE和EGFR的异常表达可能影响NSCLC患者的预后。

Abstract:Objective To investigate the expression of tumor necrosis factor-a-converting enzyme (TACE) and EGFR in primary tumors and metastases of patients with non-small cell lung cancer (NSCLC) and their prognosis. Methods Liquid chip technology, RT-PCR, Western Blot and other methods were used to measure membrane-bound tumor necrosis factor (M-TNF), TACE, EGFR level in 57 cases of NSCLC and 14 cases of benign lesions. The expressions of TACE, EGFR were compared, between NSCLC patients and healthy people, and between primary and metastatic lesions. Results The M-TNF and TACE abnormal expression rate was 66.67% and 64.91%, EGFR positive expression rate was 77.19% in 57 cases of NSCLC; TACE over-expression was not found in all 14 cases of benign lung lesions; The expressions of TACE and EGFR in NSCLC metastases were significantly higher than those in the primary tumor, which had a positive correlation with prognosis. Conclusions Detection TACE and EGFR expression in NSCLC can determine the degree of malignancy, invasion and metastasis valid reference index, TACE and abnormal expression of EGFR may influence the prognosis of patients with NSCLC.
