摘    要:目的 探讨livin在非小细胞癌(NSCLC)组织中的扩增与表达,分析该基因在NSCLC发生、发展过程中的意义.方法 对在2005至2010年收集的NSCLC的石蜡包埋组织标本,采用荧光原位杂交(FISH)和免疫组织化学(IHC)的方法,检测livin基因的表达情况.结果 经IHC检测,50例NSCLC组织中livin表达阳性率为82.00%,而癌旁组织和肺良性病变组织中livin低表达(6.67%和5.00%),差异有统计学意义(P<0.05).而经FISH检测,livin在NSCLC中表达阳性率为88.00%,两种检测方法的结果无明显差异(P>0.05).结论 FISH和IHC两种方法在检测NSCLC组织中livin基因表达具有一致性.Livin在NSCLC组织中特异性高表达,可能与NSCLC的发生、发展密切相关,有望成为NSCLC诊疗的新靶点.

Abstract:Objective To investigate the amplification and expression of livin in non-small cell carcinoma (NSCLC) tissue, and analyze the significance of livin in the occurrence and development process of NSCLC. Methods The paraffin-embedded tissue specimens of NSCLC from 2005 to 2010 were used to detect the expression of livin gene by fluorescence in situ hybridization (FISH) and immunohistochemistry (IHC). Results The positive expression rate of 50 cases of NSCLC tissues by IHC was 82.00%, but the adjacent tissues and benign lung lesions showed low expression of livin (6.67%, 5.00%); there was statistical difference (P 〈0.05). The positive expression rate of 50 cases of NSCLC tissues by FISH was 88.00%, which had no statistical difference compared with IHC (P 〉0.05). Conclusions FISH and IHC are consistent in the detection oflivin gene expression in NSCLC tissue. The specific high expression of livin in NSCLC tissue may be associated with the occurrence and development of NSCLC, which is expected to be a new target for treatment of NSCLC.
