hHGF基因肾脏电转染对大鼠肾缺血再灌注损伤的保护作用

摘    要:目的: 使用肾脏直接基因电转染方法对大鼠肾脏进行hHGF基因转染,观察该基因转染对肾脏缺血再灌注损伤的保护作用. 方法: 雄性SD大鼠20只,随机分为转染组和对照组,每组10只. 在热缺血前3 d将含有hHGF基因的质粒用电转染方法转入大鼠肾脏,3 d后建立大鼠肾热缺血模型,观察hHGF的药代动力学,并对缺血肾脏进行功能和组织学评价. 结果: hHGF基因电转染组肾组织及血浆hHGF基因表达水平高于对照组(P<0.01). 转染组血肌酐水平低于对照组(P<0.01),肾功能恢复快于对照组. 组织学评价显示,转染组肾小管细胞凋亡评分低于对照组(P<0.05). 转染组淋巴细胞、巨噬细胞及中性粒细胞浸润均较对照组少. 结论: 肾脏hHGF基因电转染组hHGF药代动力学和治疗效果均显示,该基因电转染对肾脏缺血再灌注损伤有保护作用.

Abstract:AIM:To investigate the protective effect of direct electrotransfer of hHGF gene against renal ischemia-reperfusion injury.METHODS:Male SD rats(n=20)were randomly divided into study group and control group,10 in each group.The plasmid of hHGF gene was transferred into the kidney of the rats in study group with electrotransfer technique 3 d before renal warm ischemia.The pharmacokinetics of hHGF and the function and histological change of the kidney after ischemia-reperfusion injury were determined.RESULTS:The expression of hHGF gene in the renal tissue and the plasma of the animals in study group was significantly higher than those in controls(P<0.01).The serum creatinine level in study group was significantly lower than controls(P<0.01)and the rats in study group had faster recovery of renal function when compared to controls.It was shown by histological evaluation that there were less apoptosis scores of tubular cells and less lymphocyte,macrophage and neutrophil infiltration in study group as compared to controls(P<0.05).CONCLUSION:The direct kidney electrotransfer of hHGF gene into the kidney is effective in preventing renal ischemia-reperfusion injury in rats.
