急性脑梗死患者血清中细胞因子表达的抗体芯片分析

摘    要:目的 应用细胞因子抗体芯片技术分析急性脑梗死患者不同时段血清中细胞因子表达的变化。 方法 收集急性脑梗死40例,按不同发病时段分为发病2 h组、4 h组、6 h组、24 h组及72 h组,每组8例;另外,健康对照组8例。采用细胞因子抗体芯片技术平台,测定血清中60种细胞因子。 结果 与健康对照组比较脑梗死组患者血中细胞因子表达谱发生了明显变化:①有15种细胞因子表达水平普遍呈现为高表达,包括:IL-1β、IL-1ra、IL-2、IL-5、IL-6、IL-7、IL-10、IL-15、IL-16、GM-CSF、M-CSF、TNF-α、LIGHT、GDNF、Leptin等。其中,有部分在脑梗死发病早期表达高于后期,包括:IL-6、IL-7、IL-10;另有一部分在后期表达高于早期,包括:IL-5、GM-CSF;还有部分在脑梗死的某时段特别高,包括:IL-5、IL-3、IL-15、MCP-4;②有9种细胞因子表达水平普遍呈现为低表达,包括:BMP-4、Eotaxin-3、IGFBP-3、IGFBP-4、GCP-2、I-309、MCP-1、RANTES、CNTF。 结论 细胞因子可能直接或间接参与炎症细胞的活化和浸润,并在神经元的损伤与修复过程中起重要作用。

Expression of serum cytokines in patients with acute cerebral infarction detected by antibody chip

Abstract:Objective To analyse the expression of cytokines in serum from patients who are in the different periods of acute cerebral infarction by the cytokines antibody chip technology. Methods Fourty cases of acute cerebral infarction patients were collected,according to different onset time,divided into five groups,2 hours,4 hours,6 hours,24 hours and 72 hours,8 cases in each group.In addition, 8 healthy cases were used as controls.Apply cytokines antibody chip technology to detect 60 kinds of cytokines in serum. Results Compared with healthy controls,the expression of cytokines in blood spectrum of cerebral infarction patients has changed significantly.①There are 15 kinds of cytokines generally appears as a high level,including:IL-1β,IL-1ra,IL-2,IL-5,IL-6,IL-7,IL-10,IL-15,IL-16,GM-CSF,M-CSF,TNF-α,LIGHT,GDNF,Leptin and so on.Among them,some are higher in the early cerebral infarction incidence,including:IL-6,IL-7,IL-10.Otherwise some express more in late than early,including:IL-5,GM-CSF.Some are particularly high in one specific period of cerebral infarction,including:IL-5,IL-3,IL-15,MCP-4.② There are 9 kinds of cytokines expressing lower level,including:BMP-4,Eotaxin-3,IGFBP-3,IGFBP-4,GCP-2,I-309,MCP-1,RANTES,CNTF. Conclusion The cytokines may directly or indirectly involve in the activation and infiltration of the inflammatory cells,and play an important role in the process of the damaging and repairing the neurons.
