摘    要:目的:考察黄芪提取物对IgAN患者IgA1异常糖基化的作用及其分子机制研究。方法:本研究通过分离培养IgA肾病患者和非IgA肾病对照者的外周血淋巴细胞,运用免疫组化结合分子生物学手段,研究黄芪提取物对IgA1异常糖基化的调节作用及其作用机制。结果:在IgAN患者的外周淋巴细胞上清液内,黄芪提取物组的IgA1异常糖基化程度显著降低(P<0.05),Cosmc mRNA表达水平显著提升(P<0.05)。结论:黄芪提取物在体外实验中表现出降低IgAN患者IgA1异常糖基化程度的能力,其机制可能与之调节Comsc的基因表达有关。

Research of the regulation effect of Astragalus extract on abnormal glycosylation of IgA1 of lgA nephrosis patient and its mechanism
Abstract:Objective: To investigate the effect of Astragalus extract on abnormal glycosylation of IgA1 in IgAN patients and its mechanism. Methods: In this study, peripheral blood lymphocytes in IgAN and non-IgAN patients were isolated and cultured, and we tried to found out whether and how Astragalus extracts could regular abnormal glycosylation of IgA1 by immunohistochemistry and molecular biology. Results: In IgAN patients, the degree of abnormal glycosylation of IgA1 in Astragalus extracts group reduced significantly (P<0.05), while Cosmc mRNA levels rose significantly (P<0.05). Conclusion: In vitro experiments, Astragalus extracts could reduce the degree of abnormal glycosylation of IgA1 in IgAN patients, which might probably have something to do with increasing Comsc gene expression.
