呼吸道合胞病毒M2-1基因表达对人肺腺癌PAa细胞生物学特性的影响

摘    要:目的观察呼吸道合胞病毒(RSV)M2-1基因的表达对人肺腺癌PAa细胞生物学特性的影响。方法①采用四甲基偶氮唑蓝(MTT)法测定细胞生长曲线和不同浓度丝裂霉素(MMC)作用下细胞的存活率;②采用流式细胞仪(FCM)、DNA凝胶电泳、透射电镜技术观察细胞凋亡。结果PAa/M2-1、PAa细胞的倍增时间分别为2.79和3.24d,PAa/M2-1细胞较PAa细胞生长速度快。不同浓度MMC作用下,转染后PAa/M2-1细胞的存活率均明显低于PAa细胞;PAa细胞和PAa/M2-1细胞的凋亡发生率分别为0.58%和5.6%;DNA凝胶电泳PAa/M2-1细胞可见清晰的DNA梯状条带;透射电镜观察到PAa/M2-1有凋亡细胞。结论M2-1基因的表达可促进PAa细胞生长,对不同浓度丝裂霉素表现为敏感性增加,并有自发性凋亡发生,以上结果为进一步研究RSV与肺癌的关系提供了有力证据。

Abstract:Objective To observe the influence of the expression of respiratory syncytial virus M_~2-1 gene on the biology characteristics of human lung adenocarcinoma PAa cell. Methods The cell's growth curve and the chemosensitivity on MMC were observed by MTT. The apoptosis cell were also observed by the use of electron microscopy, flow cytomemetry and DNA gel electrophoresis. Results The growth time of PAa and PAa/M_~2-1 cell was 2.79 days and 3.24 days respectively. The alive proportion of PAa/M_~2-1 cell was lower than PAa cell remarkably.The apoptosis percentage of PAa and PAa/M_~2-1 cell was 0.58% and 5.6% respectively. The clear DNA ladder were observed in the DNA gel electrophoresis and the apoptotic PAa/M_~2-1 cell were also observed in cultural cell by using the TEM. Conclusion The expression of M_~2-1 gene can promote the cell growth and enhance the chemosensitivity on MMC. The difference was significant (P<0.01). It can also induce apoptosis of PAa/M_~2-1 cell to some extent. These results may provide more detailed information to support the association between RSV infection and lung cancer development.
