Abstract:Objective To investigate the effects of novel Isoflavones on the prevention and treatment of high glucose-induced cataracts.Methods Eight-week-old male Wistar rats were randomly divided into the normal group(n=10,given 0.3%Carboxymethyl cellulose),control group(n=10,given 10%high-concentration fructose),low-dose Isoflavones group(n=9,given novel Isoflavones 1.0 mg·kg-1·d-1)and high-dose Isoflavones group(n=9,given novel Isoflavones 10 mg·kg-1·d-1).The systolic and diastolic blood pressures were determined by blood pressure meter.The blood glucose was determined by blood glucose meter,and the vision was examined by ophthalmoscope.The morphological changes of the lens were recorded by photography.The rat lenses were taken,and the Ellman assay,hydrogen peroxide decomposition method,thiobarbituric acid reaction,colorimetric determination,and nitro blue tetrazole reduction method were used to measure the levels of reduced glutathione(GSH),lens catalase(CAT),lipid peroxidation(MDA),glutathione peroxidase(GSH-Px),and superoxide dismutase(SOD),respectively.Results Starting from the 4th week,the systolic blood pressure,diastolic blood pressure,and blood glucose levels in the control group significantly increased compared with those in the normal group(all P<0.05).Compared with the control group,the systolic blood pressure,diastolic blood pressure,and blood glucose levels significantly decreased in the low-dose and high-dose Isoflavones groups(all P<0.05).The 4 groups of rats developed cataracts at the end of 8 week.Compared with the control group,the incidence of stage 3 and 4 cataract formation was significantly reduced in the low-dose and high-dose Isoflavones groups,respectively(P<0.05,P<0.01).Compared with the control group,the lens opacity was significantly improved in the low-dose and high-dose Isoflavones groups(all P<0.05).The levels of GSH,GSH-Px,CAT,SOD and MDA in the low-dose and high-dose Isoflavones groups were significantly higher than those in the control group(all P<0.05).Conclusion The novel Isoflavones may prevent the formation of cataracts by reducing the lens oxidative stress in patients with hypertension and diabetes.
