摘    要:目的基于16S rDNA技术研究高良姜、红豆蔻、大高良姜3味山姜属中药黄酮类成分对胃溃疡寒证大鼠肠道菌群的影响,探讨药用植物"亲缘关系-微生态-调节作用"之间的相关性。方法制备大鼠胃溃疡寒证模型,模型成功后,随机分为阳性组,高良姜组,红豆蔻组和大高良姜组,连续给药2 d,末次给药12 h后采集大鼠粪便,提取肠道菌群DNA,于Hiseq2500 PE250测序平台进行16S rDNA V3-V4区测序分析。结果 (1)高良姜组、红豆蔻组和大高良姜组的菌群结构很大程度上具有相似之处,其中红豆蔻组与大高良姜组菌群结构具有更高的相似性。(2) 3味山姜属中药黄酮类成分可改善胃溃疡寒证模型大鼠肠道菌群的紊乱,增加肠道菌群的丰度与多样性,降低埃希氏-志贺菌属、拟杆菌属等有害菌的丰度,提高乳酸杆菌属、普雷沃氏菌属等有益菌的丰度。(3)起到抗胃溃疡寒证作用的指示物种可能是普氏菌属、双歧杆菌属、乳酸杆菌属和分段丝状菌属。结论 (1)3味山姜属中药黄酮类成分对胃溃疡寒证大鼠肠道菌群的影响与亲缘关系相关,红豆蔻与大高良姜的亲缘关系相关性更密切。(2) 3味山姜属中药黄酮类成分发挥抗胃溃疡寒证的作用与调控肠道...

Abstract:Objective Based on 16 S rDNA technology to study the effect of flavonoids from the three kinds of Alpinia herbs, Gaoliangjiang(Alpinia officinarum Hance), Hongdoukou(Alpinia galanga Will) and Dagaoliangjiang[Alpinia galangal(L.)Wild] on the intestinal flora of rats with gastric ulcer of cold syndrome. To discuss the correlation of "phylogenetic relationship-microecology-regulation" of medicinal plants. Methods The rat gastric ulcer of cold syndrome model was prepared. After the model was successful, they were randomly divided into positive group, Gaoliangjiang(Alpinia officinarum Hance) group, Hongdoukou(Alpinia galanga Will) group and Dagaoliangjiang[Alpinia galangal(L.)Wild] group. The rats were given continuous administration for 2 d. 12 hours after the last administration, the feces were collected and extracted. The DNA of intestinal flora was sequenced and analyzed on the Hiseq2500 PE250 sequencing platform for 16 S rDNA V3-V4 region. Results(1)The flora structure of the Gaoliangjiang(Alpinia officinarum Hance) group, Hongdoukou(Alpinia galanga Will) group and Dagaoliangjiang[Alpinia galangal(L.)Wild] group were similar to a large extent, and the flora of the Hongdoukou(Alpinia galanga Will) group and Dagaoliangjiang[Alpinia galangal(L.)Wild] group had higher similarities.(2)The flavonoids of three kinds of Alpinia herbs can improve the disturbance of intestinal flora in rats with gastric ulcer of cold syndrome, increase the abundance and diversity of intestinal flora, reduce the abundance of harmful bacteria such as Escherichia-Shigella and Bacteroides, and increase the abundance of beneficial bacteria such as Lactobacillus and Prevotella.(3)The indicator species that play a role in resisting the cold syndrome of gastric ulcer may be Prevotella, Bifidobacterium, Lactobacillus and Candidatus Arthromitus. Conclusion(1)The influence of the flavonoids of the three kinds of Alpinia herbs on the intestinal flora of rats with gastric ulcer of cold syndrome is related to the phylogenetic relationship, and the phylogenetic relationship between Hongdoukou(Alpinia galanga Will) and Dagaoliangjiang[Alpinia galangal(L.)Wild] is more closely related.(2)The flavonoids of three kinds of Alpinia herbs have an anti-cold syndrome effect of gastric ulcer and are closely related to the regulation of intestinal flora.
