糖尿病患者白内障房水晚期糖基化终末产物及转化生长因子的变化

摘    要:目的检测糖尿病患者白内障前房液中晚期糖基化终末产物(advancedg lycation end products,AGEs)与转化生长因子-β(transforming growth factor,TGF-β)及葡萄糖的含量,探讨AGE、TGF-β2水平与糖尿病患者白内障的内在联系。方法收集糖尿病患者白内障、老年性白内障的前房液,采用生化方法、酶联免疫吸附测定(enzyme—linked immunosorbent assay,ELISA)法对48例糖尿病患者白内障和54例老年性白内障的房水中葡萄糖、AGEs、TGF-β2的含量进行检测。结果糖尿病合并白内障患者房水中葡萄糖水平和AGEs含量分别为(6.83±1.40)mmol/L、(355.79±195.64)pg/mL,显著高于老年性白内障患者(4.74±1.30)mmol/L、(161.67±86.23)pg/mL],两组间差异具有统计学意义(P〈0.01),TGF-β2在糖尿病患者白内障的房水(368.00±264.23)pg/mL较老年性患者(732.00-4-394.57)pg/mL明显降低,两组间差异具有统计学意义(P〈0.01)。结论糖尿病患者白内障房水中葡萄糖和AGEs含量显著高于老年性白内障,而TGF-β2的含量则降低。AGEs和TGF-β2在糖尿病患者白内障房水中含量的变化提示两者可能与糖尿病患者白内障的发生、发展有关。(中国眼耳鼻喉科杂志,2008,8:368—370)

Abstract:Objective To investigate the levels of glucose advanced glycation end-products (AGEs) and transforming growth factor beta( TGF-β ) in the anterior chamber of the diabetic cataract patients and evaluate their roles in diabetic cataract. Methods The aqueous humor of 48 cases of the diabetic cataract patients and 54 cases of age-related cataract patients were extracted as the experimental group and the control respectively. The content of the glucose was detected by biochemical methods, and the AGEs and TGF-β2 were determined using enzyme-linked immunosorbent assay (ELISA) technique. Results The concentration of glucose and AGEs in aqueous humor in diabetic cataract patients (6.83 ± 1.40) mmol/L, (355.79 ± 195.64) pg/mL] was significantly higher than that in the age-related cataract patients (4.74 ± 1.30) retool/L, (161.67 ± 86.23 ) pg/mL)] while the concentration of TGF-β was significantly lower in diabetic (368 ± 264.23 ) pg/mL] cataract patient than that in the control (732.00 ± 394.57 ) pg/mL ]. Conclusions The levels of glucose and AGEs increased in the aqueous humor of diabetic cataract patients in comparison with the age-related cataract. However, TGF-β2 in the aqueous humor of diabetic cataracts patients decreased obviously. These results indicated that AGEs and TGF-β might play an important role in the pathogenesis of diabetic cataract. ( Chin J Ophthalmol and Otorhinolaryngol,2008,8:368-370)
