IL-1β对牙龈成纤维细胞和牙周膜细胞上ICAM-1表达调节的免疫组化研究

目的 :了解牙龈成纤维细胞 (humangingivalfibroblast,HGF)、牙周膜细胞 (periodontalligamentfibroblast,PDLF)上细胞间粘附分子 1(intercellularadhesionmolecule- 1,ICAM - 1)的表达以及白细胞介素 - 1β(interleukin -1β ,IL - 1β)作用后ICAM - 1的表达。 方法 :取正畸拔牙 ,体外培养牙龈成纤维细胞和牙周膜细胞 ,检测其未受和受IL - 1β作用后ICAM - 1的表达情况 ,图像分析结果。结果 :正常牙龈成纤维细胞、牙周膜细胞上ICAM - 1表达阴性或弱阳性 ,IL - 1β作用后 ,ICAM - 1表达强阳性 ,和对照组相比 ,有显著性差异 (P <0 .0 1)。 结论 :牙龈成纤维细胞、牙周膜细胞受IL - 1β作用后ICAM - 1的表达增强 ,提示ICAM - 1参与牙周炎的病理过程。

The immunohistochemical study of interleukin-1 to Regulate Intercellular adhesion molecule-1 expression on cultured human gingival fibroblasts and periodontal ligament fibroblasts

Objective To investigate intercellular adhesion molecule-1 (ICAM-1) expression on human gingival fibroblast (HGF) and periodontal ligament fibroblast (PDLF) and the effects of interleukin-1β to regulate intercellular adhesion molecule-1 expression on HGF and PDLF.Methods Using cell culture and immunohistochemical S-P method to observe the expression of ICAM-1.Results The level of ICAM-1 expression in IL-1β treated HGF and PDL fibroblasts were statistically significant ( P <0.01) compared to normal untreated controls.Conclusions This study demonstrates that IL-1β upregulates ICAM-1 expression in both HGF and PDLF,suggesting that ICAM-1 is involved in the pathogenesis of periodontal disease.
