摘    要:采用地高辛标记探针,建立了一种快速检测和筛选转入肾素基因小鼠的方法。对实验中杂交液的选择、杂交液中探针的浓度,预杂交时间,抗体孵育前封闭时间和洗膜条件等影响因素进行了系统观察与分析,从而确定了以地高辛标记探针检测转入肾基因小鼠方法中的最佳实验条件。结果表明,该方法不仅安全,快速,而且灵敏度高,稳定性好。

Abstract:Transgenic mice carrying human renin gene were generated by means of pronucleus injection:Screening for transgenic mice was carried out by Southern-blot hybridization using digoxigenin(DIG ) labeled probe. We inquired some factors influencing detection such as selection of hybridization buffer, concentration of probe in hybridization buffer, the length of time of prehybridization and blocking before antibody binding and condition of washing step. Finally, we elected the optimal method to detect transgenic mice carrying human renin gene. The tests show that this method is not only safe, quick, but aslo sensitive and steady.
