miR-223在肾透明细胞癌患者外周血中的表达及其临床意义

目的探讨肾透明细胞癌(ccRCC)患者外周血miR-223的表达及其与临床病理参数的关系。方法收集该院ccRCC患者(n=40)术前及术后7 d外周血标本为实验组,门诊体检健康人群(n=80)外周血标本为对照组。利用实时定量聚合酶链式反应(RT-PCR)仪器检测miR-223在实验组与对照组中的表达水平,使用受试者工作特征(ROC)曲线评价血浆miR-223诊断ccRCC及判断其预后的价值。收集ccRCC患者的临床数据,探讨外周血中miR-223表达水平与患者病理特征的关系。结果实验组患者外周血中miR-223表达量明显高于对照组(P<0.05);实验组患者术后7 d较术前外周血中miR-223的表达量已明显减少,但仍高于对照组(P<0.05);且ccRCC患者外周血中miR-223的表达水平与肿瘤TNM分期、肿瘤分化程度、肿瘤大小有关(P<0.05)。结论ccRCC患者外周血中miR-223呈高表达,且与其肿瘤TNM分期、肿瘤分化程度、肿瘤大小有关,术后其表达水平降低,可用于ccRCC的诊断和评估病情进展。

Expression and clinical significance of miR-223 in peripheral blood in patients with renal transparent cell carcinoma

【Objective】To investigate the expression of peripheral blood miR-223 in patients with clear cell renal cell carcinoma(ccRCC)and its relationship with clinical pathological characteristics.【Methods】The experiment was designed according to the 1:2 case control study,in which the blood samples of ccRCC patients(n=40)were collected before and 7 days after surgery as the experimental group,and the peripheral blood samples of the health-checked population(n=80)were collected as the control group.Using real-time quantitative polymerase chain reaction(RT-PCR)instrument to detect the expression level of miR-223 in the experimental group and control group,the receiver operating characteristic(ROC)curve was used to evaluate the value of plasma miR-223 in diagnosing ccRCC and determining its prognosis.The clinical data of ccRCC patients were collected to explore the relationship between miR-223 expression level in peripheral blood and pathological characteristics of patients.【Results】MiR-223 was significantly higher in the peripheral blood of ccRCC patients than in the healthy population,and its level was significantly reduced in 7 days after surgery(P<0.05),and the expression level of miR-223 in peripheral blood in ccRCC patients was related to tumor TNM staging,tumor separation,tumor size,with statistical significance(P<0.05).【Conclusion】MiR-223 in peripheral blood in ccRCC patients was highly expressed,and was related to tumor TNM staging,tumor separation,tumor size,and its expression level was significantly reduced after surgery,which can be used for diagnosis and evaluation of the progression of ccRCC.
