睾酮对高脂饲养小鼠主动脉超微结构的影响

摘    要:目的研究睾酮对高脂刺激的雄性小鼠主动脉超微结构的影响。方法56只小鼠随机分为4组,其中1、2、3组给去势术,4组行假手术。2组(小剂量补充睾酮组)、3组(大剂量补充睾酮组)分别给予10mg.kg-1.d-1、15mg.kg-1.d-1的睾酮腹腔注射;1组(低睾酮组)、4组(正常对照组)给予安慰剂。各组小鼠均高脂饲养,3个月后测定部分血小板、血脂、凝血指标,并电镜观察主动脉的超微组织结构变化。结果1组、3组光镜及电镜下均呈早期动脉粥样硬化(AS)改变,1组与2组、4组相比,LDL-C、PAI明显升高,HDL-C下降,P<0.05;各组之间血小板计数、血小板聚集率、TCH、TG差异均无显著性,P>0.05。结论内源性睾酮有防止AS的作用,适量补充外源性睾酮可以替代内源性睾酮的作用,低血清睾酮及大量补充睾酮均促进AS的发生。

Abstract:Objective To study the effect of endogenic and ectogenic testosterone(T) on artery pathology in cholesterol-fed male mice.Methods 56 mice were divided randomly into four groups.Group 1 was castrated only(hypotestosterone,n=14),group 2(n=14) and group3(n=14) were given respectively 10mg.kg-1.d-1,15mg.kg-1.d-1 T through abdominal cavity injection after castration,placebo were given to group 4(n=14,sham castration)and group1.All rats were fed with cholesterol-rich diet during 12-week treatment period.When the test was over,blood samples were obtained for detection of platelet count,platelet aggregation rate,TC,HDL-C,TG,LDL-C,PAI,then mice were killed and thorax arteries were taken out for examinations of pathology by optical and electronic microscope.Results Early period AS appeared in arteries of group 1 and 3.Compared with group 2 and 4,both LDL-C and PAI in group 1 were obviously higher,while HDL-C was significantly decreased(P<0.05),PAI was higher in group 3 than in group 4(P<0.05).No deference was observed in each group in platelet count,plateletaggregation rate,TC and TG.Conclusion Endogenic T plays a role in inhibiting aortic AS in male mice,supplying proper dose ectogenic T can take place of the role of endogenic T.Hypotestosterone and high dose T can expedite the development of AS.
