摘    要:目的 探讨高迁移率族蛋白-1(HMGB1)对增生性瘢痕成纤维细胞(HSFbs)的调控作用.方法 增生性瘢痕组织取自中山大学附属第一医院烧伤科行瘢痕整复术的患者,组织块法培养HSFbs.通过细胞增殖、实时荧光定量PCR(qRT-PCR)、迁移和胶原网收缩实验检测不同浓度(0~200 ng/mL) HMGB1对HSFbs的影响.结果 1.56、3.13、6.25、12.50、25.00 ng/mL HMGB1呈剂量依赖方式刺激HSFbs的增殖,HMGB1对HSFbs Toll样受体4(TLR4)、α-滑肌肌动蛋白(α-SMA)、非肌肉肌球蛋白Ⅱ(NMMⅡ)mRNA的表达有促进作用而对基质金属蛋白酶1-α (MMP1-α)无,HMGB1对HSFbs迁移及收缩有促进作用但可被TLR4阻断剂抑制.结论 HMGB1可能通过TLR4调控HSFbs而在增生性瘢痕的形成过程中起作用.

Abstract:Objective To investigate the regulatory effects of high mobility group box 1 (HMGB1)on cultured hypertrophic scar fibroblasts (HSFbs).Methods Hypertropic scar tissue was obtained from patients undergoing scar plasty at Department of Burns Surgery,the First Affiliated Hospital,Sun Yat-Sen University.Fibroblasts of scar tissue were cultured by tissue block method.The second passage of cells were collected.Effects of HMGB1 (0 ~200 ng/mL) on cultured HSFbs were measured by cell proliferation,qRTPCR,migration and HSFbs-populated collagen lattice assay.Results HMGB1 stimulated cell proliferation with a dose-dependent increase at concentration 1.56,3.13,6.25,12.50,25.00 ng/mL.HMGB1 could increase Toll like receptor 4 (TLR4)、α-smooth muscle actin (α-SMA)、nonmuscle myosin Ⅱ (NMM Ⅱ1)mRNA expression but had no effect on matrix metalloproteinase 1-α (MMP1-α).The positive effect of HMGB1 on migration and contraction of HSFbs could be inhibited by TLR4 blocking antibody.Conclusion The results have suggested that HMGB1 may exert its function on HSFbs and the formation of hypertrophic scar through TLR4 signaling.
