大孔树脂纯化丹参总酚酸的研究

摘    要:目的:确立大孔树脂纯化丹参总酚酸的最佳纯化工艺。方法:以静态吸附率和解吸率为考察指标,确定大孔树脂型号,并确定纯化工艺参数。结果:选择HPD-400型号大孔树脂用于丹参总酚酸的纯化工艺,最佳吸附工艺条件为:上样吸附流速为2BV/h,上柱液的浓度为0.5 g生药/mL,pH值为4;解析前,以3BV的水洗脱除杂,最佳解析工艺条件为:选择乙醇为洗脱剂,洗脱剂浓度为40%,pH值为5,控制解析流速为3BV/h。结论:所建立的方法简便、准确,可用于丹参总酚酸的纯化。

Abstract:Objective: To establish the purification technology of Total phenolic acid of Danshen by macroporous resin. Method: Static adsorption rate and desorption rate were taken as indexs to optimize the type of macroporous resin and purification technology parameters. Result: The HPD-400 macroporous resin was chosen in the purification technology of Total phenolic acid of Danshen. Optimum adsorption technological conditions were as following: The adsorption rate was 2BV/h. The concentration was 0.5 g raw drug/mL with pH of 4.3BV water was used to elute impurity before desorption. Optimum desorption technological conditions were as following: Eluted with 40% ethanol aqueous at the flow rate 3BV/h, and pH was 5. Conclusion: The method is simple, accurate, specific and can be applied in the purification of Total phenolic acid of Danshen.
