代谢综合征大鼠模型的建立与评价及增龄因素的影响

摘    要:目的建立、评价喂养型代谢综合征(MS)大鼠动物模型,观察其大血管组织病理变化,探索增龄因素对其影响。方法将青年和老年SD雄性大鼠随机分为青年正常组,老年正常组,青年模型组、老年模型组,各15只。采用高脂、高糖、商盐饲料喂养制备模型。分别于8、16w测定各组动物的空腹血糖(FPG)、空腹胰岛素(FINS)、血脂和0,8,12,16时血压。观察16w时各组动物颈动脉、冠状动脉、主动脉组织病理变化和内驻脂肪系数。结果与青年正常组比较,青年模型组和老年模型组大鼠胰岛素敏感性指数均明显下降(P〈0.01);总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和TC/HDL-C明显升高(P〈0.05,P〈0.01)。除青年正常组外各组动物血压较0w时均有所升高(P〈0.05,P〈0.01);老年模型组的上述各项指标改变均势较青年模型组更为明显。模型组大鼠的失血管内膜损伤明显,平滑肌细胞增生,其中老年模型组大鼠的血管损伤较青年模型程度严重。结论喂养复合模型饲料可以制备出符合或接近临床的MS大鼠模型。增龄因素对MS及其大血管损伤具有重要影响。

Abstract:Objective To establish and evaluate a new rat model of metabolic syndrome and to investigate effect of ageing on it.Methods The young and old rats were randomly divided into young control group,old control group,young model group and old model group,15 ones every group.The model was fed by high lipid, sugar and salt diet.FPG,FINS and serum lipid(TC,TG,LDL-C,HDL-C) levels and systolic blood pressure(SBP) were measured at the 8th and 16th weeks.Pathological changes of carotid artery,coronary artery,aorta and bowel fat coefficient were observed at 16th week.Results The insulin sensitivity index decreased(P<0.01),the levels of serum lipid(TC,TG,LDL-C and TC/HDL-C) increased obviously in young and old model group(P<0.05,P<0.01)than those of young control group.Except young control group,the levels of SBP in others were stepped up(P<0.05,P<0.01).The above indexes in old model group had obvious trend than those of young model group.Rats in model group had obvious great vessel histopathology and proliferated SMC,while vessel injury in old model group were more serious than that of young model group.Conclusions The complex model diet feeding could build rat model of metabolic syndrome(MS) according with or approaching to clinic practice.Ageing factor should play an important role in MS and its great vessel impairment.
