硫化氢对代谢综合征小鼠血糖和血胰岛素水平的调节

摘    要:目的 观察硫化氢(H2S)对代谢综合征小鼠血糖和血胰岛素水平的影响及可能机制.方法 8~10周的雄性C57BL/6J小鼠10只作为正常对照组(CON组),同周龄雄性KK/Upj-Ay/J小鼠随机分为MS+NaHS组和MS组,每组各6只,MS+NaHS组每日腹腔注射0.25 mL H2S供体NaHS(78 μmol/kg),CON组和MS组每日腹腔注射等量生理盐水,均连续干预4周.采用ONE TOUCH Ultra血糖仪测定血糖值,放射免疫分析法测定胰岛素值,荧光显微镜观察小鼠C2C12骨骼肌细胞株对2-脱氧荧光葡萄糖(2-NBDG)的摄取能力.结果 KK/Upj-Ay/J小鼠的腹围、Lee指数和空腹血糖、胰岛素以及三酰甘油、总胆固醇、低密度脂蛋白胆固醇均比CON组显著升高(P< 0.01);MS+NaHS组小鼠空腹血糖和胰岛素比MS组分别降低24.2%和46.98%(P< 0.05);NaHS干预后,骨骼肌摄取葡萄糖增加(P<0.05);在高糖(16.7 mmol/L)刺激下,不同浓度NaHS干预后,胰岛素分泌水平下降(P<0.05).结论 NaHS可以促进骨骼肌细胞对葡萄糖的摄取,同时可以直接抑制高糖刺激下的胰岛素分泌.

Abstract:Objective To explore the effect of hydrogen sulfide on fasting blood glucose and serum insulin in MS mice and its possible mechanism.Methods C57BL/6J mice (8-10 weeks) were selected in the control group (CON group,n=10).KK/UpjAy/J mice (8-10 weeks) were randomly divided into the MS + NaHS group (n=6) and MS group (n=6).MS + NaHS group was given an injection of 0.25 mL NaHS (78 μmol/kg) daily for 4 weeks.The CON group and MS group were given an injection of 0.25 mL saline daily for 4 weeks.The fasting blood glucose level was determined by ONE TOUCH Ultra and the serum insulin level was determined by radioimmunoassay.2-NBDG uptake of C2C12 was measured by fluorescence microscope.Results The abdomen circumference,Lee's index,FBG,insulin level,TC,TG,LDL-C of KK/Upj-Ay/J mice were all higher than these in CON group (P 〈 0.01).Compared to MS group,the FBG and insulin level in MS+NaHS group reduced to 24.2%and 46.98% (P 〈 0.05).2-NBDG uptaked of C2C12 increased in MS+NaHS group (P 〈 0.05).Insulin secretion levels dropped on the effect of different levels of NaHS under the stimulate of 16.7 mmol/L glucose (P 〈 0.05).Conclusion NaHS can advance the 2-NBDG uptake of C2C12,but restrain the insulin secretion levels under the stimulate of high glucose.
