| 摘 要： | 目的探讨c-kit蛋白和mRNA在慢传输性便秘结肠的表达.方法乙状结肠标本取自12例慢传输性便秘患者和8例年龄相匹配的非梗阻性结直肠癌患者.用Western blot 方法检测c-kit蛋白表达,用RT-PCR方法检测mRNA表达情况.结果聚丙烯凝胶电泳结果显示c-kit蛋白质一145×103的条带,与对照组相比,慢传输性便秘结肠c-kit蛋白表达明显下降(P＝0.021);RT-PCR产物经琼脂糖凝胶电泳显示c-kit基因和β-actin(内参照)两条带,经半定量分析发现c-kit mRNA表达降低(P=0.029).结论慢传输性便秘乙状结肠c-kit蛋白和mRNA表达均明显降低,提示c-kit基因表达下调与慢传输性便秘Cajal间质细胞减少有关.

| Abstract: | Objective To examine the expressions of c kit mRNA and c kit protein in the colon of patients with slow transit constipation (STC). Methods The sigmoid colon samples from 12 STC patients and 8 age matched patients with nonobstructed colorectal cancer were used in this study. Expression of c kit protein was detected by Western blot analysis, and expression of c kit mRNA was detected by RT PCR. Results Compared with those of controls, the c kit mRNA and protein expressions were significantly declined in the colon of slow transit constipation ( P <0.05). Conclusion Significant downregulation of c kit gene and protein expressions may play an important role in the decreases in interstitial cells of Cajal in slow transit constipation. |
