头针对大鼠局灶性脑缺血再灌注后脑组织Ang-1 mRNA的影响

摘    要:目的观察头针治疗对大鼠局灶性脑缺血再灌注后脑组织内血管生成素(Ang)-1 mRNA表达的影响。探讨其变化规律并分析脑缺血后Ang-1在缺血性脑卒中发病中的作用机制。方法大鼠随机分为正常对照组、假手术组、模型组和治疗组,治疗组又分为缺血再灌注后6、24、48、96h组,采用改良线栓法制作永久性大脑中动脉闭塞模型,RT—PCR技术检测各组大鼠脑组织缺血半影区内Ang-1 mRNA的表达水平。结果正常对照组、假手术组Ang-1 mRNA中等量表达,与模型组比较,头皮针组各时间点大鼠脑组织Ang-1 mRNA含量增多,其中脑缺血再灌注48hAng-1 mRNA含量达高峰,与模型组比较,差异有统计学意义(P〈0.05或0.01)。结论Ang-1可能在局灶性脑缺血后与其他血管特异性生长因子共同参与缺血半影区的新血管形成.有利于缺血半影区脑组织功能恢复。

Expressions of Angiogenin-1 mRNA after Scalp Acupuncture in Focal Cerebral Ischemia in Rats Brain Tissues

Abstract:Objective: To observe the scalp acupuncture in treatment of cerebral blood vessel generation reperfusion of rats with focal cerebral ischemia in the expression changes of different time points and to explore the changing rules and analysis the action mechanism of Ang-1 after cerebral ischemia in cerebral arterial thrombosis.Methods: Rats were randomly divided into the normal control group ,the control group,the model group and the treatment group. The treatment group was divided into the ischemia reperfusion group at 6,24,48 and 96 h, with the improved wire plug do legal permanent middle cerebral artery occlusion (MCAO) model. And then, some indicators were used RT-PCR to detect each group rats ischemic penumbra area Ang-1 mRNA expression level. Results: In the normal control group,control group of Ang-1 mRNA of the equivalent expression,compared with model group,Ang-1 mRNA of the scalp needle group rats group were increased at each time point. Compared with the model group, the cerebral ischemia reperfusion 48 h that was the highest concentration of these levels was higher significantly (P〈 0.05 or 0.01). Conclusion: That Ang-1 is imoportant after focal cerebral ischemia and other vascular specific growth factor in ischemic penumbra formation of new blood vessels and inhibit apoptosis of neurons is conducive to improve the ischemic penumbra area blood supply and promote nerve functional recovery.
