RP-HPLC法同时测定白英中绿原酸及咖啡酸的含量

目的建立RP-HPLC法同时测定白英药材中绿原酸、咖啡酸含量的方法。方法色谱柱为Di-amonsil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数0.05%的磷酸水溶液(体积比25∶75),检测波长327 nm,柱温45℃,流速1.0 mL.min-1。结果绿原酸质量浓度在0.5~32.0 mg.L-1(r=0.999 9)内与峰面积呈良好的线性关系,平均回收率为99.9%,RSD为1.52%。咖啡酸质量浓度在10.0~640.0μg.L-1(r=0.999 9)内与峰面积呈良好的线性关系,平均回收率为99.9%,RSD为1.35%。结论该方法简便、准确、重复性良好,可用于白英药材的质量控制。

Simultaneous determination of the contents of chlorogenic acid and caffeic acid in Solanum lyratum Thunb. by RP-HPLC

Objective To establish an HPLC method for simultaneous determination of the contents of chlorogenic acid and caffeic acid in Solanum lyratum Thunb. Methods The operation was carried out on Diamonsil-C₁₈ column (250 mm×4.6 mm, 5 μm) with the mobile phase consisting of a mixture of methanol-0.05% phosphoric acid (V∶V =25∶75). The flow rate was 1.0 mL·min^-1, UV detection wavelength was set at 327 nm and the column temperature was 45 ℃. Results The calibration curve of chlorogenic acid was linear in the range of 0.5-32.0 mg·L^-1 (r=0.999 9). The mean recovery of chlorogenic acid was 99.9%, RSD was 1.52%. The calibration curve of caffeic acid was linear in the range of 10.0-640.0 μg·L^-1(r=0.999 9). The mean recovery of caffeic acid was 99.9% and RSD was 1.35%. Conclusions The HPLC method is found to be simple, accurate, sensitive and reproducible. It can be used for the determination of the contents of chlorogenic acid and caffeic acid in Solanum lyratum Thunb.
