摘    要:目的 探讨IL-6在多囊卵巢综合征(polycystic ovary syndrome, PCOS)高胰岛素血症和胰岛素抵抗中的作用。方法 在体外培养的大鼠胰岛细胞培养液中加入10、100和1000pg/ml IL-6,于培养24、48、72h后,采用放射免疫法检测IL-6对胰岛细胞胰岛素分泌的影响。结果 培育48h时,1000pg/ml IL-6板孔中的胰岛素浓度高于对照组,差异有统计学意义(P<0.05);培育72h时,100、1000pg/ml IL-6板孔中的胰岛素浓度均高于对照组,差异有统计学意义(P<0.05)。结论 IL-6在体外具有刺激胰岛细胞分泌胰岛素的作用,这可能与PCOS患者常出现高胰岛素血症、胰岛素抵抗现象有关。

Abstract:ObjectiveTo investigate the effect of interlukin-6 (IL-6) on insulin secretion in rat pancreatic islet cells in vitro. Methods Primarily cultured pancreatic islet cells were treated with IL-6 at concentration of 10,0 and 1000pg/ml, respectively. The insulin contents in culture supernatant were detected by radioimmunoassay (RIA) at 24,8 and 72h after culture. ResultsThe insulin levels in culture supernatant with IL-6 1000pg/ml at 48h after culture were significantly higher than those in controls (P<0.05). The insulin levels in supernatant with IL-6 100pg/ml and 1000pg/ml were significantly higher than those controls at 72h after treatment. Conclusion IL-6 can induce insulin secretion in pancreatic islet cells in vitro, which may be associated with hyperinsulinemia and insulin resistance involved in polycystic ovarian syndrome.
