氟化钠、亚砷酸钠对大鼠肝细胞增殖的影响

摘    要:目的研究氟化钠(NaF)和亚砷酸钠(NaAsO2)单独及联合作用对大鼠BRL-3A细胞增殖的影响。方法设NaF(1.0×10-5~1.0×10-1)mol/L、NaAsO2(1.0×10-7~1.0×10-3)mol/L不同浓度组(组间10倍差),分别作用于大鼠肝细胞,采用MTT法测定OD值,观察其对细胞增殖的影响。NaF在1.0×10-3mol/L与NaAsO2在1.0×10-6mol/L、1.0×10-5mol/L、1.0×10-4mol/L浓度下,两者联合作用于大鼠肝细胞时,采用2×2的析因试验分析其对细胞增殖的影响。结果氟、砷单独作用对肝细胞增殖的抑制率均随染毒剂量的增大而逐渐升高,而联合组在NaF为1.0×10-3mol/L与NaAsO2为1.0×10-6mol/L、1.0×10-5mol/L、1.0×10-4mol/L浓度时,其抑制率均明显低于单独组,边际轮廊均数图显示两线段交叉,两毒物的联合作用呈拮抗作用。结论氟、砷化合物对大鼠肝细胞增殖呈抑制作用,两者联合作用时表现为拮抗作用。

Effect of NaF and NaAsO2 on proliferation of rat hepatocyte

Abstract:Objective To study the effect of NaF and NaAsO2 on proliferation of BRL-3A rat cells.Methods Rat hepatocytes were used to evaluate proliferating effects of NaF and NaAsO2 at concentrations of(1.0×10-5-1.0×10-1) mol/L and(1.0×10-7-1.0×10-3) mol/L(10 times difference between two consecutive groups),proliferation cell was observed by OD values measured by MTT assay.The combined effects of NaF at concentrations of 1.0×10-3 mol/L and NaAsO2 at 1.0×10-6 mol/L,1.0×10-5 mol/L,1.0×10-4 mol/L on proliferation of rat hepatocyte were evaluated by 2×2 factorial analysis.Results Fluorine and arsenic had inhibitory effect on proliferation of rat hepatocyte.Proliferation inhibition rate increased with the exposure dose increase.However,the combined effects of NaF at concentrations of 1.0×10-3 mol/L and NaAsO2 at 1.0×10-6 mol/L,1×10-5 mol/L,1.0×10-4 mol/L were all antagonistic,inhibition rate was obviously inclined separate group.Two line segments were crossed in mean contour diagram.Conclusion The proliferation of hepatocyte is inhibited with Fluoride and arsenic exists in rat,the combined effects show antagonism.
