摘    要:摘要]目的研究参麦注射液对新生大鼠缺氧缺血性脑损伤(HIBD)的干预作用。方法建立新生鼠HIBD模型。免疫组化方法检测缺氧缺血(HI)后2h、12h、24h、3d、7d、14d各时间点新生大鼠右侧大脑海马CAl区Bax、Bcl-2蛋白表达情况。实验各观察点随机分为假手术组(s组)、参麦注射液治疗组(SM组)、生理盐水对照组(c组),每纽9只。结果Hl后2h,SM组、c组右侧大脑海马CAl区Bcl-2蛋白表达及Bax蛋白表达均已有升高,24h达到高峰,14d各组降到与SM组同一水平:SM组、c组在2h、12h、24h、3d、7ct点与s组比较均有显著性差异,在14d与s组比较无显著性差异;SM组在24h、3d、7d与c组比较,Bcl-2蛋白表达较c组高,在2h、12h、14d点与c组比较无显著性差异。SM组在12h、24h、3d、7d与c组比较,Bax蛋白表达较c组降低,在2h、14d与c组比较无显著性差异。结论参麦注射液可以提高抑制凋亡基因Bcl-2的表达及降低促凋亡基因Bax的表达。

Effects of Shenmai injection on hypoxic-ischemic brain damage of neonatal rats

Abstract:Objective It is to investigate the intervention effect of Shenmai injection on neonatal rats with hypoxic-ischemic brain damage. Methods Neonatal rat models with HIBD were established. Immunohistochemistry was used to detect the ex- pression of Bax and Bcl - 2 in the hippocampal CA1 region of right brain in neonatal rats with HIBD at 2 h, 12 h, 24 h, 3 d, 7 d, 14 d after hypoxie-ischemic (HI) insult. Every observed spot was separated into three groups: sham operation (S) , con- trol group (C) and Shenmai injection treatment (SM). Results The expression of Bcl -2 in right hippocampal CA1 region in group SM, group C at 2 h after HI insults had already began to increase, peak at 24 h and then gradually decreased. The ex- pression of Bcl -2 in right hippocampal CA1 region in every group had no difference at 14 d (P 〉 0.05 ). The expression of Bcl - 2 in right hippocampal CA1 region in group SM at 24 h, 3 d, 7 d was significantly more than that of group C( P 〈 0.05, P 〈 0.01 ). The expression of Bcl - 2 in right hippocampal CA1 region in group SM and group C had no difference at 2 h, 12 h, 14 d (P 〉 0.05 ). The expression of Bax in right hippocampal CA1 region in group SM, group C at 2 h after HI insults had already began to increase, peak at 24 h and then gradually decreased. The expression of Bax in right hippocampal CA1 region in every group had no difference at 14 d (P 〉0. 05 ). The expression of Bax in right hippocampal CAl region in group SM at 12 h, 24 h, 3 d, 7 d was significantly less than that of group C(P 〈0.05, P 〈0.01 ). The expression of Bax in right hipp- ocampal CA1 region in group SM and group C had no difference at 2 h, 14 d(P 〉0.05). Conclusion Shenmai injection can significantly enhance the expression of Bcl - 2 in right hippocampal CA1 region and decline the expression of Bax in right hippocampal CA1 region.
