刺五加对大鼠迟发性脑血管痉挛脑组织细胞凋亡的影响

摘    要:目的探讨刺五加在防治蛛网膜下腔出血(SAH)、迟发性脑血管痉挛(DCVS)及其所致的迟发性脑缺血中的作用机制.方法"枕大池二次注血"建立大鼠SAH-DCVS动物模型,采用TUNEL法和流式细胞术动态观察刺五加对大鼠SAH-DCVS脑组织细胞凋亡的影响.结果TUNEL检测发现刺五加组在SAH后随着给药时间的延长和给药次数的增加,与同期单纯注血组比较海马、皮质、基底节区及血管内皮细胞、血管平滑肌细胞可见凋亡细胞明显减少,有明显的药物时间依赖性;流式细胞仪检测亦发现刺五加组凋亡率明显降低,与相对应单纯注血组比较均有明显差异(P<0.001).结论刺五加可以通过抑制细胞凋亡,防治SAH-DCVS及其所致的迟发性脑缺血、迟发性神经细胞损伤.

Abstract:Objective To discuss the mechanism of acanthopanax senticosus to prevent and cure SAH-DCVS and related late response brain hemorrhage.Methods To construct rat SAH-DCVS model with"the methods of two-times blood injection in occipital cerebral fossa supplement".To utilize TUNEL and Flow Cytometry to observe the affect of acanthopanax senticosus on rat SAH-DCVS brain organization cell apoptosis.Results The number of apoptosis cells of AS group were obviously decreased in hippocampus, cortex, basilar section district, vessel endothelial,and smooth muscle cell. With the prolonging of time and the frequency of using medicines after SAH ,compared with the pure injection group at the same time ,we observed that the number of apoptosis cells were obviously decreased in AS group(P<0.001)and the AS group had obvious time dependence by ways of TUNEL and Flow Cytometry.Conclusion Acanthopanax senticosus can prevent and cure SAH-DCVS and delayed related nerve fiber damage by restraining cell apoptosis.
