灌肠方对溃疡性结肠炎大鼠血清IL-4和IL-10的影响

目的建立溃疡性结肠炎大鼠模型,研究灌肠方对溃疡性结肠炎大鼠的治疗作用及对血清IL-4和IL-10的影响,探讨灌肠方防治溃疡性结肠炎的作用机理。方法将60只SD大鼠随机分为正常组,空白对照组,中药低、中、高剂量组和美沙拉嗪组,共6组,每组10只。应用2,4,6-三硝基苯磺酸(TNBS)复制溃疡性结肠炎动物模型,灌肠方低、中、高剂量组分别给予不同剂量灌肠方灌胃,美沙拉嗪组予以美沙拉嗪灌胃,正常组及空白对照组灌服等体积蒸馏水。18天后观察大鼠结肠大体形态学及组织病理学改变,采用ELISA法检测各组大鼠的血清IL-4和IL-10的水平。结果灌肠方能有效提高IL-4和IL-10的水平,并对损伤的大肠粘膜有明显的修复作用。结论灌肠方对大鼠实验性溃疡性结肠炎疗效显著,促进抗炎因子IL-4和IL-10的产生和表达可能是其治疗机制之一。

Effect of Guanchangfang to the Ulcerative Colitis Rats Serum Levels of IL-4 and IL-10

Objective To study the effect of Guanchangfang on the ulcerative colitis repairment and rats serum levels of IL-4 and IL-10. Methods 60 SD rats were randomly divided into normal group,model group,Guanchangfang low,middle and high dose group,Mesalazine group,10 in each group. Ulcerative colitis rats model were copied by 2,4,6-TNBS. The general morphological and histopathological changes were observed and the rat serum levels of IL-4,IL-10 were detected by ELISA 18 days later. Results Guanchangfang effectively improved the serum levels of IL-4 and IL-10,and had significant effect of repairing the intestinal mucosal injury. Conclusion The significant effect of Guanchangfang on rats experimental ulcerative colitis may be attributed to raising the expression of IL-4 and IL-10.
