p53抑癌基因蛋白过表达在肉瘤发生中作用的研究

目的 探讨不同类型人肉瘤中p53抑癌基因蛋白的表达。方法 对44例不同类型肉瘤石蜡组织中p53蛋白的表达进行研究，同时以免疫组化染色作对照。结果 44例不同类型肉瘤中21例显示p53蛋白表达阳性，表达率为48％，其中平滑肌肉瘤（67％）、骨肉瘤（50％）和滑膜肉瘤（47％）中表达率较高，而良性肿瘤（除1例局部细胞生长活跃区呈现阳性外）常呈阴性染色。原位免疫PCR染色阳性率高于免疫组化染色结果，染色强度前者也明显高与后者。结论：p53在人自发性肉瘤中有较高的突变率，提示可能参与人肉瘤的发生。原位免疫PCR显示了较好的定位性、敏感性和重复性。

EFFECTS OF OVEREXPRESSION OF p53 PROTEIN IN OCCURRENCE OF HUMAN SARCOMAS

Objective To investigate the overexpression of p53 protein in different kinds of human sarcomas. Methods Expression of p53 protein were observed in 44 cases of sarcomas by using immuno-PCR in situ and contrasted with immunohistochemistry staining.Result the result showed 48%(21/44) sarcomas were positive for p53 protein, which were 67% in smooth muscle sarcomas, 50% in osteosacomas, 47% in synovial sarcomas. But benigin tumors were negtive. The rate of positive staining for immuno-PCR were higher than immunohistochemistry and positive intensity of the former higher thanthe latter. Conclusion The rate of p53 mutation were higher in spontaneous human sarcomas and indicated it can be role in occurrence of sarcomas. Immuno-PCR showed higher sensitivity, favorable location and repetition.
