细胞外基质成分异常与获得性肾囊肿大鼠肾小管间质损害的关系

摘    要:目的探讨细胞外基质(ECM)成分异常与获得性肾囊肿大鼠肾小管间质损害的关系。方法建立化学诱导的获得性肾囊肿疾病(ARCD)大鼠模型。采用免疫组织化学检测不同损害级别的小管间质ECM及α-平滑肌肌动蛋白(SMA)的表达。应用RT-PCR方法检测其mRNA的表达,并分析两者之间的相关性。结果ARCD组不同损害级别的小管间质肾脏组织中,胶原(Col)Ⅰ、Ⅲ、纤连蛋白(FN)及α-SMA表达均显著增加,与小管间质的损害程度呈正相关;层粘连蛋白(LN)的表达组间比较无显著差异,与小管间质的损害程度亦无显著相关性。结论ECM成分异常导致其结构重塑,可能参与了ARCD的形成过程。

Abstract:Objective To explore the relationship between abnormal expression of extracellular matrix (ECM) and the impairments of tubulointerstitium in rats with acquired renal cystic disease (ARCD). Methods Immunohistochemistry was used to detect the protein expression of ECM and a-SMA in tubulointerstitium of rats with ARCD, and the mRNA expression was detected by RT-PCR. According to different degree of impairments, the rats were divided into 4 groups. Correlation between the abnormal expression of extracellular matrix and the impairments of tubulointerstitium was also examined. Results In these 4 groups, the expression levels of Col I , Col III, FN or a-SMA were different, and were significantly and positively correlated with the degree of tubulointerstitial impairments, but the expression of LN did not show the same characteristics as above. Conclusion The abnormal changes of Col I , Col III, FN and a-SMA can lead to reconstruction of ECM, which may be involved in the development of ARCD.
