脾虚证大鼠肝、脾组织蛋白激酶C活性变化的比较研究

摘    要:目的探讨脾气虚证、脾阳虚证和脾阴虚证大鼠肝、脾组织蛋白激酶 C(PKC)活性变化的异同。方法采用饮食不节、疲劳过度和药物损伤等复合因素建立脾气虚证、脾阳虚证和脾阴虚证大鼠模型;采用底物磷酸化法检测 PKC 活性。结果脾气虚证、脾阳虚证和脾阴虚证模型脾组织细胞膜 PKC 活性明显降低(P<0.01),但组间比较差异无显著性(P>0.05);脾组织细胞浆和肝组织细胞膜(脾气虚证模型组除外)以及肝组织细胞浆 PKC 活性明显升高(P<0.01),除脾阴虚证模型与脾阳虚证模型脾组织细胞浆及脾气虚证模型与脾阴虚证模型肝组织细胞膜 PKC 活性无明显差异外,其余每两组之间均存在明显差异(P<0.01)。补脾中药对脾组织细胞浆、肝组织细胞膜和细胞浆 PKC 活性均有明显的降低作用(P<0.01),尤以温运脾阳及滋补脾阴药作用显著。结论脾气虚证、脾阳虚证和脾阴虚证模型肝、脾组织细胞 PKC 活性变化存在一定的差异;补脾中药能够调节脾虚证大鼠肝、脾组织 PKC 活性。

Abstract:Objective To study the differences and similarities of protein kinase C(PKC)activity in liver and splenic tissues among rats with various types of Pi-deficiency syndromes(PDS)as Pi-Qi deficiency syndrome(PQD),Pi-yang deficiency syndrome(PyaD)and Pi-yin deficiency syndrome(PyiD).Methods Rat models of various PDS types were established by such complex factors as improper diet,over fatigue and drug damage.PKC activity was detected by substrate phosphorylation method.Results PKC activity in cell membrane of splenic tissue significantly decreased in all the rats with PDS(P<0.01),showed no significant difference among different types(P>0.05).But PKC in cytoplasm of splenic tissue and PKC in both cell member(except PQD)and cytoplasm of liver tissue was significantly increased(P< 0.01).Except that between PyiD and PyaD in cytoplasm of splenic tissue,and between PQD and PyiD in cell membrane of liver tissue,comparison of PKC activity between different types all showed significant difference(P<0.01).Chinese drugs for supplementing Pi,especially that for warmly promoting Pi-yang and nourishing Pi-yin,showed significant effect in reducing PKC in splenic cytoplasm,liver cell membrane and cytoplasm(P<0.01).Conclusion PKC activities in liver and splenic tissues of different types of PDS showed definite differences,Chinese drugs for supplementing Pi could regulate the PKC activity in liver and splenic tissues in rats.
