温肾生精饮治疗生精障碍的实验研究

摘    要:目的:临床上利用温肾生精饮治疗男性生精障碍,已取得了显著疗效,本实验利用生精障碍小鼠模型探讨温肾生精饮促进精子生成的可能作用机制。方法:利用环磷酰胺对昆明小白鼠建立生精上皮损伤模型,对造模后的小鼠分别灌服温肾生精饮或等量生理盐水,灌服20天后检测小鼠附睾尾精子质量,并进行睾丸组织学检查和超微结构观察。结果:中药组小鼠附睾活精子的百分率和精子密度显著高于模型组;中药组小鼠睾丸生精小管结构规则,生精上皮排列整齐,上皮层数较模型组显著增多;中药组小鼠睾丸的超微结构与模型组相比,间质细胞胞质内脂滴较少,线粒体较多,溶酶体较多,间质细胞显示出旺盛的分泌能力。结论:温肾生精饮提高了间质细胞的分泌功能,维持着睾丸生精小管内高水平睾酮,以增加生精上皮层数,促进精子发生,提高正常精子的比率。

Abstract:Objective: The WenShenShengJing decoction is effective in the clinic for the spermatogenesis obstruction. To further study the mechanism of these Chinese Medicines promoting spermatogenesis. Methods : The mouse model of spermatogenesis obstruction was made with cyclophosphamide, and then these model mice were treated with WenShenShengJing decoction for 20 days as Chinese Medicine group, while the model group treated with equivalent saline at the same time. After treatment, the quality of epididymal sperm and testicular structure were observed. Results : The viability and number of sperm were significantly higher in the Chinese Medicine group than those in model group. Compared with model group, the lumina of the seminiferous tubales in the Chinese Medicine group was regular with more spermatogenic epithelia, and had more sperm. Electron microscopy showed that Leydig cells in the Chinese Medicine group had less lipid droplet, more mitochondria and lysoomes than that in model group. Conclusions : The WenShenShengJing decoction can significantly enhance the secretory function of Leydig cells to maintain the testosterone level in the seminiferous tubules, increasing the layers of spermatogenic epithelia and facilitating normal spermatogenesis.
