知母水溶性多糖的分离、纯化及初步研究

摘    要: 目的分离、纯化出知母水溶性多糖,研究其性质。方法粗多糖经反复冻融S、evage法脱蛋白、高速离心、超滤、柱色谱等方法分离纯化得级分AAB1。经HPLC、比旋光度测定等方法,证明AAB1为均一组分。用气相色谱分析多糖组成。结果AAB1由Xyl,Man,Gal,GalA,Glc组成,其摩尔比为1∶11∶1.75∶7.92∶1.2。平均相对分子质量为5.47×104。结论AAB1为酸性均一杂多糖。

Abstract:OBJECTIVE To study the composition and characterization of water-soluble crude polysaccharide AAB1 from Anemarrhena asphodeloides Bge.METHODS The crude polysaccharide was freezed-thowed and deproteinized by Sevage method repeatedly,fractionated by ultrafiltration and gel chromatography Sepharose on CL-6B column,so AAB1 was obstained.Its homogeneity was proved by HPLC and specific optical rotation measurement etc.The composition of AAB1 was studied by means of GC etc.RESULTS AAB1 was made up of Xyl,Man,Gal,GalA and Glc with the molarity rate of 1∶11∶1.75∶7.92∶1.2 and its molecular weight was about 5.47×104.CONCLUSION AAB1 is a acid heteropolysacchride.
