近交系大鼠心脏移植模型建立的实验研究

摘    要:目的:建立近交系大鼠同种异位腹腔心脏移植动物模型。方法:采用改良的ono术式建立Brown Norway到Lewis大鼠的腹部异位心脏移植,将供心升主动脉、主肺动脉分别与受体腹主动脉、下腔静脉进行端侧吻合。采用分别结扎右上腔和下腔静脉后一次性集束结扎法取心,行端侧血管吻合连续缝合法。结果:共进行了48例大鼠心脏异位移植,经过预实验16只的技术总结,正式实验32只手术成功率为96.88%,供心冷、热缺血时间和心脏复跳时间明显缩短,手术时间减少至60min。结论:加强供心的综合保护和围手术期的处理,是移植模型建立的关键因素。

Abstract:Objective: To establish the model of inbred strain rat heart heterotopic transplantation.Methods:Male Lewis rats were used as recipients and Brown-Norway rats as donors,and the heterotopic transplants were done using ono's method with some modifications.The ascending aorta and pulmonary artery of donor were anastomosed end-to-side to the recipient abdominal aorta and inferior vena cava respectively.Results:48 heart transplantations were performed.We summarized surgical technique after sixteen rats of preliminary experiment.The successful rate was improved to 96.88% in 32 rats of formal transplantation.Time of cold ischemia,hot ischemia and rebeat were shorten significantly and operational time reduced to 60 min.Conclusion:To strengthen the protection of donor heart and perioperative treatment is the key point in the heart heterotopic transplantation in inbred strain rat.
